Sindora glabra is one of National Second Class Key Protected Wild Plant,belonging to genus Sindora of Caesalpiniaceae.As a rare oil-secreting species with high economic value,it can be utilized for biodiesel,drugs,spices and high-quality wood,indicating with the great development potential.However,the germplasm resources of S.glabra have been increasingly endangered due to the increasing fragmentation of habitats and the weak self-renewal ability of itself.Therefore,it is urgent to strengthen the conservation of germplasm resources of S.glabra.It has become an important measure of conserving germplasm resources to screen out excellent germplasms for breeding and utilization by carrying out genetic marker analysis and genetic diversity evaluation.In this study,10-year-old S.glabra germplasm conservation forest in Shaoping Forest Farm,Pingxiang City,Guangxi Province was taken as the research object.Genetic variation of leaf phenotypic traits was revealed,the specificity of SSR loci information was elaborated,and genetic structure and variation were explored through the analysis of leaf phenotypic traits,the development and exploitation of SSR markers based on transcriptome,in an attempt to provide scientific basis for strengthening the conservation and creation of germplasm resources of S.glabra.The main results of this study were as follows:(1)There were significant differences in leaf phenotypic traits among different populations of S.glabra indicating abundant variations among populations;Among the leaf quality traits of S.glabra,the fluctuation range of color difference was 11 grades,where leaf color was the most abundant and petiole color was the most stable.Leaflets were mostly oval in shape,while compound leaves were mostly ellipse in shape.Under this test condition,the most fluctuating character was the area of the leaflets,and the most stable character was the leaf shape index of the lobule.The population fluctuated most widely in leaf quantitative traits was LD3,and the differences of leaf traits among the remaining 10 populations was not significant.The correlation analysis among the leaf traits showed that the leaf length of compound leaves was positively correlated with the remaining traits,and the length ofcompound leaves was extremely significantly positively correlated with other traits.Relative variation degree of compound leaf traits was greater than that of lobule.Based on the variation of leaf quantitative traits of S.glabra,clustering results showed that the genetic diversity of leaf quantitative traits was not closely related to geographical distribution.(2)A total of 283,998 Unigenes were obtained from S.glabra based transcriptome splice,and 97443 SSR loci were found,with the occurrence frequency of 34.31%,indicating abundant SSR loci in the transcriptome of S.glabra and short distribution distance between different loci.SSR in the transcriptome of S.glabra was also relatively abundant in category and quantity.Selecting the DNA from the leaves of S.glabra as the samples,11 pairs of SSR primers were screened for PCR amplification,and 94 alleles were amplified with 0.588 of mean value of polymorphic information loci.Generally,the SSR loci polymorphism in the transcriptome of S.glabra belongs to the upper-middle type.(3)The genetic variation and structure of S.glabra germplasm resources were analyzed by 11 polymorphic sites..The genetic variation of S.glabra germplasm resources was mainly intra-population variation.The mean values of Na,Ne,I,Ho,He,and F in populations were8.545,2.81,1.071,0.535,0.583,and 0.098,which indicated that the genetic diversity was high.The value of gene flow fluctuated widely among different loci,with an average value of 4.685,indicating more frequent gene exchanges and less genetic differentiation among different populations of S.glabra.Eleven populations could be divided into four categories by UPGMA clustering mothod based on genetic identity,which had a low coincidence with geographical sources.
