Broussonetia papyrifera is perennial deciduous trees or shrubsis.The entire tree of Broussonetia papyrifera has high application value in various fields.It can assist in the treatment of various diseases,and can also improve the soil ecological environment,absorb heavy metals in the soil,provide raw materials for feed and substrates,and also be the raw materials for papermaking.However,the current material supply of Broussonetia papyrifera is severely in short supply.The traditional breeding technology has little effect and the survival rate of cutting propagation is low.Therefore,many materials need to be imported from abroad to meet domestic demand.The use of tissue culture technology can effectively improve the reproduction efficiency,shorten the planting cycle,and effectively solve the problem of shortage of raw materials.In this experiment,the tissue culture seedlings of Broussonetia papyrifera were used as explants,using hormones such as CPPU,6-BA,KT,NAA,IAA and IBA,to explore the effects of different medium formulations,different sucrose content,different hormones and their relative concentration ratios,two-step rooting method,dipping rooting method,high-concentration NAA treatment on different days and activated carbon on the rooting of Broussonetia japonica.Screen the best rooting treatment through the above experiments,in order to provide an important basis for production practices and the demand for seedlings of framing trees and subsequent research.The test results are as follows:1.The best medium suitable for rooting of Broussonetia papyrifera is 1/2MS medium,the average root length is 2.13cm,the average number of roots is 2.27,and the rooting rate is 46.7%.2.The optimum amount of sucrose added to the rooting medium of Broussonetia papyrifera is 30g/L,the average root length is 2.23cm,the average number of roots is 2.26,and the rooting rate is 50.0%.3.With different auxins and relative concentrations,the optimal rooting medium for constitutive trees was 1/2MS+NAA 0.1 mg/L,the average root length is 2.49cm,the average number of roots is 3.57,and the rooting rate is 56.7%.4.The optimal rooting medium obtained by combining auxin NAA with different cytokinins and their concentrations is 1/2MS+NAA 0.1 mg/L+CPPU 1.0 mg/L,the average root length is 3.24 cm,the average root number is 5.83,and the rooting rate is 76.7%.5.The rooting culture of Broussonetia papyrifera was carried out by the two-step rooting method,and the rooting effect was best when the concentration of NAA was 80 mg/L,the average root length is 3.48 cm,the average root number is 2.83,and the rooting rate is 66.7%.6.Add 80 mg/L of NAA to the culture medium,and perform rooting culture on different days.The results show that the treatment has the best effect for 8 days,the average root length is 3.27 cm,the average root number is 2.43,and the rooting rate is 69.0%.7.The rooting culture of Broussonetia papyrifera was carried out by dipping method.The NAA concentration is 100 mg/L,the dipping time is 60min,the rooting is the best,the root length is 3.22cm,the number of roots is 2.73,and the rooting rate is 75.9%.8.The amount of activated carbon most suitable for Broussonetia papyrifera rooting is 0.1%,the root length is 2.64cm,the number of roots is 3.03,and the rooting rate is 63.3%.9.The experiment was repeated three times with 20 treatments in each group.After 20 days,the survival rate was counted as 40%,25%and 30%respectively,and the average transplanting survival rate was 31.67%.
