Haemonchus contortus is a kind of gastrointestinal nematode which has the greatest impact on ruminants.The emergence of drug resistance of Haemonchus contortus seriously threatens the development of animal husbandry.In order to screening the genes related to the drug resistance of albendazole in Haemonchus contortus,for resistance mechanism research and clinical drug resistance strain identification,this study based on the transcriptome data,get the SNP information of the sensitive strain and albendazole resistant strain of Haemonchus contortus,analyzed the function of enrichment,The 4 differential genes were screened,for bioinformatics analysis and PCR sequencing verification.The single nucleotide polymorphisms at positions 167,198 and 200 of the isotype-1 ?-tubulin gene in albendazole resistant populations collected from Uxin Qi,Qahar Youyi Houqi,Horqin Youyi Qianqi in Inner Mongolia were investigated.The detection of P450,GST and ABC gene expression under the stimulation of different drug concentrations of Haemonchus contortus albendazole resistant strains was tested.The results are as follows:?1?A total of 6090 differential genes of Haemonchus contortus HC-WSrBZl and HC-WSrBZ2 were enriched on 50 GO Term.The 11 significant enrichment pathways were screened out.Hc-CYsBZ and Hc-CYrBZ have a totall of 6757 differential genes enriched on 50 GO Term.The 19 significant enrichment pathways were screened out.A total of 208 differential genes with SNP sites were enriched on 26 GO Term in Hc-ssBZ and Hc-XArBZ.The 10 significant enrichment pathways were screened out.The 4 differential genes HCON00192020,HCON 00037800,HCON00129090,and HCON00178650 screened out have a clear difference between the albendazole resistant strains and the sensitive strains,and can be used as molecular markers for drug resistance.?2?The detection of the isotype-1 ?-tubulin gene of the population of albendazole in the three regions of Uxin Qi,Qahar Youyi Houqi,Horqin Youyi Qianqi in Inner Mongolia revealed that there was no mutation at position 167,and There are mutations at 198 and 200 sites.In general,the mutation frequency of 198 sites is higher than that at 200 sites.4 genotypes were found in 3 regions,namely rr-198 and SS-200,SS-198 and rr-200,SS-198 and rS-200,rS-198 and rS-200.?3?Through q-PCR detection,under the stimulation of different drug concentrations,the expression levels of the three genes P450,GST and ABC were significantly different,which initially proved to be related to drug resistance.
