In this experiment,"JinChun NO.4" cucumber was selected as scion and "yunnan black seed pumpkin" as rootstock greenhouse grafted cucumber was selected as test material.The experiment was carried out in the same greenhouse and in an independent area completely isolated from each treatment plot.The effects of high temperature and CO2 enrichment on morphology and stomatal density of cucumber leaves were studied.Effects of high temperature and CO2 enrichment on stomatal development related hormones in cucumber leaves;And the effects of high temperature and CO2 enrichment on stomatal development related genes in cucumber leaves.The main research results are as follows:1.Under the condition of CO2 enrichment and stomatal density changes of cucumber leaf morphological index of response:pure high temperature treatment of cucumber leaf thickness,fresh weight,dry weight and leaf area was significantly lower than the treatment,and extend along with the processing time,inhibition,high temperature and high temperature,CO2 enrichment can significantly relieve inhibition of cucumber in long-term processing,increase the stomatal density,promote the increase of leaf growth and dry matter.2.Under the condition of high temperature,CO2 enrichment cucumber leaf abscisic acid(ABA)and ethylene(ETH)and auxin(IAA)change of response:under the condition of normal temperature,long-term provision of CO2 causes the cucumber leaf abscisic acid content increased,the inhibition of stomatal development,but under the condition of high temperature CO2 enrichment(1000-1300 umol/l)can significantly increase cucumber leaf content of ethylene and auxin,promote stomatal development,stomata density increased.3.The high temperature,CO2 enrichment of cucumber leaf stomata related genes:the influence of high temperature,CO2 enrichment cucumber leaf EPF2 expression significantly reduce the amount of crops grown,EPFL9 and ER expression significantly increases,its SPCH express quantity is significantly higher than control during the whole processing,is significantly higher than in the treatment of the six days after high temperature and normal temperature,CO2 enrichment processing,at the same time,during the whole processing,were significantly improved the mute expression,it also promoted the stomatal density increase;The significant increase of FAMA expression in normal temperature and CO2-enriched treatment was related to the increase of SPCH and MUTE.At room temperature,the content of EPF2 in the leaves increased significantly with the addition of CO2,thus negatively regulating stomata,retardeting stomatal development and reducing stomatal density.
