Salvia miltiorrhiza,known as Danshen in China,is a perennial medicinal plant of the genus Salvia in the Labiaceae family with high medical value.The secondary metabolites in the dried roots such as tanshinone and methyltanshinonate was widely used in traditional Chinese medicine and applied in the treatment of many diseases,particularly cardiovascular disease.Because of its high medicinal value,the demand for Danshen was increasing,which led to over-exploitation of wild resource and resulted in resource shortage.Nowadays,clinical applicated Danshen mostly derived from cultivation,but continuous cultivating resulted in the chemical properties of the derived plants decreased,and virus infection also accentated the decrease degree of chemical properties.Recently,tissue culture techniques had a great development in rapid propagation,ermplasm preservation,and virus free seedlings propagation as well as genetic manipulation of medical plants.In order to improve the quality and yield of Danshen,plant tissue culture technology was used to study the propagation,and added rare earth elements to improve its quality of the optimized propagation system.At last,the contents of chlorophyll,antioxidant enzyme and secondary metabolites of the propagated plantlets were determined.The research results were as follows:Firstly,an efficient rapid propagation system of Danshen was established.The results showed that the advanced-buds induction effect was highest in MS medium containing 1.0 mg·L-1 6-benzyladenin(6-BA)and 0.1 mg·L-1(naphthaleneacetic acid)NAA.The best rooting effect is observed on MS medium adding 0.2 mg·L-1 NAA.And the survival rate of tissue culture seedlings is up to 95%.Sencondly,we investigated the effect of rare earth elements on the growth and secondary metabolism on Danshen.The seedlings were cultured for 4 weeks as explants.The rare earth elements cerium(Ce),lanthanum(La)and praseodymium(Pr)at different concentrations(50,100,200 and 300 ?M)were added to MS medium which containing 0.2 mg·L-1 NAA for root induction and culture.The results showed that the induction effect of roots in the medium with 100 ?M Pr was significantly better than Ce and La,and the total tanshinone content in the roots was 0.38 mg tanshinone IIA·mg-1·DW,an increase of 54.84%compared with the control group.At the same time,the chlorophyll contents,antioxidant enzyme activity and secondary metabolites were also increased after induced by 100 ?M Pr.In a word,adding 0.2 mg·L-1 NAA and 100 ?M Pr to MS medium can obtain higher quality Danshen tissue culture seedlings,so as to provide reference for high-quality Danshen cultivation.
