Cyclocarya paliurus is one of the endemic tree of china which has multiple efficacy in medicine, health care, timber use , and aesthetic enjoyment. Preliminary studies were complet -ed on tissue culture and regulation of secondary metabolites in this article. Principal results of the study were as follows:1. Growths of excised embryos, which were from three different seed sources ofCycloca -rya paliurus, were compared. The excised embryos were inoculated on three basic mediums: MS, WPM and improved DKW. It was found that there was a significant differences among the tree seed sources of Cyclocarya paliurus. The survivor rate (72.22%) of tissue provenances from Zhejiang provenance were higher than others. The result also showed that there were significant differences of provenances and the optimum cultural medium for excised embryos was WPM.2.It was found that MS+6-BA 4.0mg/L+IBA2.0mg/L was the optimum medium for inducing stem segment callus of Cyclocarya paliurus, MS+6-BA2.0mg/L+IBA0.5 mg/L was the optimum medium for inducing leaf explants callus of Cyclocarya paliurus, and MS+6-BA1.0 mg/L +IBA1.0mg/L was the optimum medium for callus proliferation. VitaminC was good to inhibit Cyclocarya paliurus callus from browning . The browning rate was only 5.71% .3. Zhejiang provenance has the highest inductivity of adventitious buds on the basic medium of improved DKW+6-BA2.0mg/L+IBA0.1mg/L. The inductivity was about 47.68%. Secondly was Guizhou provenance on the basic medium of improved DKW+ 6-BA 1.0mg/L +IBA0.01mg/L which the inductivity was about 10.71%.4.1/2WPM+IBA0.1mg/L+uniconazole0.5mg/L+sucrose20g/L was the best medium to ind -uce root for test tube plantlet which was subcultured at lest 4 times. The rate of rooting was affected by Subculture number and additives as well.5.Studies of Secondary metabolites regulation of Cyclocarya paliurus showed that 200 mg/L SA has the best effection on increasing the content of Isoquercitrin and Kaempferol in leaves. With such treatment, the contents were 2.28 and 1.49 times than contrast respectively; 100mg/L SA was the best treatment to Quercetin which content was 2.15 times than contrast; 400mg/L Nano-TiO2 was the best treatment to inorganic ions Fe and Ni which contents were 3.16 and 4 times than contrast respectively.
