This study used tissue separation preparation and spore separation two species of fungus separate method,4 kinds of preservation fungus methods were adopted,including glycerol-agar semi-gel method,glycerin-nutrient agar semi-gel method,inclined low temperature preservation method and fresh milk method.By observing the liquid fungus,fructification morphology,the yield of the fructification and the content of the bioactive components cordyceps polysaccharide and oxalic acid were measured.Study the effect of cordyceps militaris fungus separation and preservation methods on Cordyceps militaris growth and development and accumulation of bioactive components,the results show that:(1)Cordyceps militaris fungus can be successfully prepared by tissue separation and polyspora separation.The fungus prepared by tissue separation preparation are more stable in properties and better in quality.(2)Cordyceps militaris fructification grow 30d(days),which is the best separation period for the preservation of fungus by tissue separation preparation.The method of low temperature preservation on inclined surface is the best preservation method.Adding beef paste,peptone and other nutrients in glycerol-agar semi-gel can delay the decline of the quality of Cordyceps militaris to some extent.The liquid fungus incubate by the fungus prepared in this separation period produced fast,uniform and large numbers of fungus balls,thus the fructification of the liquid fungus incubate turned fast and deep,grew healthily,had high fructification production,with a yield of 20.71-21.86g/box.(3)when cordyceps militaris fructification grow 40d,the best separation period for the preservation of the fungus is obtained by the spore separation method.The method of low temperature preservation on the slope is the best preservation method.The addition of beef paste,peptone and other nutrients in glycerol-agar semi-gel does not delay the decline of the quality of cordyceps militaris.The liquid fungus incubate by the fungus prepared in this isolation period is ideal.Compared with other isolation periods,the liquid fungus cultured cordyceps militaris has the high fructification yield,less deformity and high fructification production,with a yield of 20.44 to 20.64g/box.(4)The content of cordyceps polysaccharide and cordyceps acid in the fructification of the fungus prepared by the tissue separation method in each separation period changed little,ranging from 4.54-6,18%and 0.203-0.381%respectively.The preparation and the separation period of the fungus had no significant effect on the content of cordyceps polysaccharide and cordyceps acid in the fructification of the fungus prepared by the tissue separation preparation.(5)The content of cordyceps polysaccharide in the fructification of the fungus in different separation periods prepared by the method of polysporosis separation varied greatly,ranging from 3.54%to 7.83%and 0.066-0.448%respectively.The content of the bioactive component cordyceps polysaccharide in the fructification by the method of inclined low temperature preservation was significantly higher than that of the other preservation methods.The method of preservation has a significant effect on the content of cordyceps polysaccharide in the fruiting body of the fungus.
