Camellia vietnamensis,as a characteristic cash crop and sunrise industrial tree species developed vigorously in Hainan Province in recent years,its flowering sterility is common in the population of Camellia vietnamensis seed trees in Hainan,and the sterility characteristics will seriously affect the production level of seed crops.it has become an important obstacle restricting the sustainable development of Camellia vietnamensis industry in Hainan.The results are as follows:Analysis of Biological characteristics and Molecular Mechanism of male sterile plants of Camellia vietnamensis in Hainan1.By comparing the external morphology of normal flowers and sterile flowers of Camellia vietnamensis in Hainan,it was obvious that the structure of male organs of sterile flowers was abnormal.In addition,there was also a significant difference in ovary morphology between normal plants and sterile plants in Hainan after flowering: the ovary of normal plants developed and expanded normally after flowering,but the ovary of sterile plants became withered and tended to fall off after flowering.From the external morphological analysis,Hainan Camellia vietnamensis is male and female sterile.2.Through the observation of pollen germination and pollen tube development characteristics of normal and sterile plants of Camellia vietnamensis in Hainan,it was found that after pollination,pollen germination and pollen tube growth of normal plants of Camellia vietnamensis in Hainan were normal.However,the pollen tube of the male sterile plant of Camellia vietnamensis in Hainan was not developed after pollination.3.The flower organ was dissected and observed by paraffin section method.the results showed that the structure of anther and embryo sac of normal plant was completely normal,but the corresponding development period of sterile plant was the same.The phenomenon of pistil and stamen abortion was obviously different from that of the normal plant: the tapetum development and degradation of the male sterile anther was abnormal,and the mature pollen grains could not be formed;the embryo sac of the sterile plant developed abnormally,and the meiosis could not be initiated or completed.4.In order to further explore the causes of sterility from a molecular point of view,transcriptome sequencing was carried out on four stages of Camellia vietnamensis normal plant and male sterile plant in Hainan using Illumina Hi Seq platform,and a total of 175,200 unigenes were obtained.However,the genes with unknown functions accounted for 34.46%,indicating that the research on the gene function of Camellia vietnamensis in Hainan is not deep enough,and the data still have the potential to be tapped.In the follow-up,the genes with unknown functions will be further excavated by means of transcriptome data and molecular markers.5.By comparing the corresponding developmental stages of normal plant and sterile plant of Camellia vietnamensis in Hainan,it was found that 6,462 genes were common differentially expressed genes in the four developmental stages of normal plant and sterile plant.In order to more accurately analyze the molecular mechanism of male and female sterility of Camellia vietnamensis in Hainan,the selected differential genes were annotated by GO and enriched by KEGG.It was found that the sterility of Camellia vietnamensis in Hainan was mainly regulated by plant hormone signal transduction pathway and metabolic pathway.6.After screening,5 differentially expressed genes related to ovule development and 5 key genes related to anther development were found.The 10 key genes screened were verified by real-time fluorescence quantitative PCR.The results of q RT-PCR verification showed that the differential multiple of gene expression was almost consistent with the transcriptome data,indicating that the differentially expressed genes obtained in this study were reliable.
