The proportion of citrus occupying Chinese fruit market is rising year by year because of its multiple species and tastes.With extended planting field,citrus needs to strengthen the adaptation to face various abiotic stress.Proline maintains plant's normal metabolism by a large amount of accumulation on adversity stress which depends on P5 CS regulating the whole proline synthesis pathway.To explore the expression model in different stresses,we cloned the gene P5CS1 in C.reshni,accompanying with bioinformation analysis,relative gene expression and functional researches such as yeast one-hybrid,subcellular location and VIGS.The results show below:1.P5CS1 protein isoelectric point is 5.29,the molecular weight is 77430.25,containing 713 amino acids and is a hydrophilic protein.From molecular cladogram,it shows that P5CS1 in C.reshni has more relative kinship with C.sinensis.Forecasting P5CS1 promoter,there are several kinds of cis-acting elements such as ABRE,MYB,RYelement which relating to drought stress,light stress and tissues specificity.2.P5 CS relative gene expression shows first up and then fall under drought,salt and low temperature stress.Analyzing P5 CS,P5CR,Pro DH gene expression for proline synthesis,P5 CS has the same expression trend in different stresses and species;P5CR in C.tangerine displays first fall and then up with stress time passing.3.Compared with other two stresses,citrus has a higher proline content under salt stress.Testing proline content on each timepoint for given stress,proline has the same changing trend and more sensitive to salt stress.C.tangerine has a higher content under drought and low temperature stress while Poncitrus trifoliata has a low but top content under salt stress.In all,P5CS1 has a precise response to salt stress in Poncirus trifoliata,C.tangerine C.reshni.The analysis for P5CS1 sequence also provides a clear direction for future study.
