The Salamandrella keyserlingii is mainly distributed in the northeast region in our country,with the relatively narrow distribution range,for which taxonomic status is relatively primitive in the tailed amphibian,and it has an important research position in the phylogenetic research of amphibians.In this paper,the method of COI gene sequence analysis and the structural characteristics of the skeletal system are used to study the genetic diversity of S.keyserlingii for 6 geographical populations,and the following conclusions were drawn:1.In the COI gene of S.keyserlingii,the average content of the four nucleotide bases was A=27.73%,T=34.67%,G=17.00% and C=20.58% respectively,the content of A+T was significantly higher than the content of G+C.The genetic distance between the six populations was ranged from 0.003 to 0.098.Among them,the distance between Tieli population in Lesser Khingan Mountains and Huma population in the Greater Khingan Mountains was smaller,shown as 0.003.There were also small genetic distance between Shangzhi Zhuziying population,Shangzhi Cuijia population,Hailin population in Zhangguangcai Peak and the Baishan population in Changbai Mountain,shown as 0.005~0.008,but the genetic distance between the population of the Greater Khingan Mountains,Lesser Khingan Mountains and Zhangguangcai Peak,Changbai Mountain was large,shown as 0.097~0.098.2.The significant genetic differentiation has occurred between the 6 populations of the S.keyserlingii.The genetic differentiation index Fst was 0.94974 among the groups,indicating that the high degree of genetic differentiation was high,and the genetic variation within the group accounted for 5.03% of the total variation.Furthermore,the variation accounted for 94.97% of the total variation,indicating that the genetic variation mainly came from the variation among populations.3.The analysis results of haplotype phylogenetic tree showed that the Tieli populations and Huma populations in the Greater Khingan Mountains and Lesser Khingan Mountains were S.keyserlingii,while the Shangzhi Zhuziying populations,Shangzhi Cuijia populations,Hailin populations and Baishan populations in ChangbaiMountain and Zhangguangcai Peak were S.tridactyla.It could be inferred that S.keyserlingii is distributed in the north of Heilongjiang Province and the northeast of Inner Mongolia in China,while S.tridactyla is mainly distributed in southern Heilongjiang Province,Jilin,Liaoning and others.4.There was no significant difference in morphological characteristics and number of bones between S.keyserlingii and S.tridactyla In the skeletal system,there were about 39 pieces of skulls,12 pieces of hyoid bones,88 pieces of limb bones,17 pieces of spine,25 pieces of tail vertebra and 34 pieces of ribs.
