Aphidius gifuensis is an excellent natural enemy for the prevention and control of Myzus persicae.In China,the technology of expanding A.gifuensis on a large scale is becoming more and more mature.However,in the practice of propagation expansion,it was found that after several generations of continuous feeding,the parasitic ability,living ability,body weight and body size of A.gifuensis all decreased to different degrees,which reduced the vitality of the population and became the key bottleneck of continuous large-scale propagation of A.gifuensis.Based on the production demand,this paper systematically studied the population degradation rule and rejuvenation technology of A.gifuensis The transcriptomic characteristics of degradation and rejuvenation were analyzed from the molecular level.The research results can enrich the basic theories of population degradation and rejuvenation of the A.gifuensis,and guide the large-scale propagation and production of natural enemies.The experimental results are as follows:1.The population degradation rule of A.gifuensisBy measuring the relationship between the degree of decline and the feeding generation,the characteristic rules of population degradation were analyzed and summarized,and the characteristics of parasitic ability,eclosion rate,sex ratio,adult life span,met thoracic tibia lengths in different generation were systematically compared,so as to define the degradation characteristics and the generation.the results show that from the F1generation to the F12generation,the parasitic ability has decreased by 67.22%and the eclosion rate has decreased by 31.91%The female rate increased by 33.32%,the adult longevity of A.gifuensis decreased by53.85%,the metathoracic tibia lengths decreased by 17.67%,metathoracic tibia lengths decreased by 28.74%,and the characteristics of the F7-F9generation declined significantly.The experiment confirmed that the 7th generation population of A.gifuensis in Breed indoors had experienced significant degeneration.2.The nutrition rejuvenation and hybrid rejuvenation of the degenerated population of A.gifuensisThe nutritional rejuvenation has a certain rejuvenation effect on the life span and parasitic ability of A.gifuensis the 10%honey water is the best,However,they are significantly lower than the hybrid rejuvenation effect;the hybrid rejuvenation has a significant rejuvenation effect on A.gifuensis The female wild population have better rejuvenation effects than the male wild population,The wild population introduced into Yunnan region has the best rejuvenation effect,and the parasitic ability can be restored to 88.21%of the F0generation.3.The transcriptome characteristics of the degeneration and rejuvenation of the aphidAccording to transcriptomics technology to analyze and compare the wild group,the degeneration group and the rejuvenation group of A.gifuensis,a total of 97654 Unigenes were obtained.During the degradation process of A.gifuensis,a total of 13,472 genes were significantly different,of which 4492 genes were up-regulated and 8980genes were down-regulated.After rejuvenation of the population of A.gifuensis,a total of 3803 gene expression levels were up-regulated and4705 gene expression levels were down-regulated.The analysis of COG annotation maps can be used to degenerate in the process,there are a large number of genes involved in energy conversion,ribosome generation,and protein modification,etc.Through the enrichment analysis of the KEGG pathway of the differential genes,it can be obtained that during the degradation process of the A.gifuensis,the Metabolic pathway Biosynthesis of other secondary metabolites pathway and Ribosome pathway have the largest number of annotated genes,indicating that these 3 pathways are more active than others,and are suitable for in-depth research.The use of GO enrichment analysis and pathway analysis on the wild group The functional analysis of EDGs in the degeneration group and the rejuvenation group showed that a large number of differential genes were enriched in the Ribosome pathway,Oxidative phosphorylation pathway,and Endoplasmic reticulum protein processing pathway,Fatty acid synthesis and Metabolism pathway,Rna degradation pathway,indicating that these pathways play an important role in the degradation and rejuvenation of A.gifuensis...
