DNA methylation plays an important role in gene expression,X-chromosome inactivation,cell differentiation,chromatin structure maintenance and embryo development.DNA methylation is also important in the regulation of ovarian activity.In this study,Guanzhong dairy goat was selected as the research object,and the whole genome DNA methylation map of ovary in estrus(ES)and estrus interval(DS)was drawn by wgbs and bioinformatics analysis.After comparative analysis,the following results are obtained:1.In the ovaries of ES and DS,the whole genome DNA methylation pattern was found.Compared with DS group,the methylation of 26910 regions was up-regulated and 21453 regions was down regulated in ES group(P ? 0.05,the change of methylation ratio ? 2).2.The analysis of differential methylation regions showed that the degree of methylation of the genomic region was low,and the degree of methylation of the upstream region connected with the genomic region was high.The methylation rate of STAR?FGF2?FGF12?BMP5?SMAD6 genes in ES samples was lower than that in DS samples(P ? 0.05,the multiple change of methylation rate ? 2).In contrast,the methylation rates of EGFR?TGFBR2?IGF2BP1 and MMD2 genes in ES group were higher than those in DS group.3.223 differentially methylated genes were found in GnRH signaling pathway(ko04912),ovarian steroidogenesis pathway(ko04913),estrogen signaling pathway(ko04915),oxytocin signaling pathway(ko04921),insulin secretion pathway(ko04911)and MAPK signaling pathway(ko04010).In this study,the high-resolution DNA methylation of oestrus(ES)and oestrus interphase(DS)ovaries in dairy goats was compared on a large scale.The results showed that NR5A2?STAR?FGF2and BMP5 genes may have potential application value in regulating oestrus behavior of dairy goats.
