The color of plant seedcoat is formed by the deposition of many anthocyanins.The biosynthesis pathway of plant pigment is relatively conservative,which is closely related to the enzyme of flavonoid biosynthesis pathway.The color of Mungbean seedcoat is an important morphological marker of Mungbean.At present,the research on the color of Mungbean seedcoat is not deep enough.Therefore,the research on the color gene of Mungbean seedcoat is of great significance to the molecular research and breeding practice of Mungbean.In order to continue to explore the genetic law of Mungbean seedcoat color,this experiment made up different combinations of Mungbean seedcoat color,and carried out statistical analysis on the phenotype of F1 and F2 generations;At the same time,the markers linked to the color of the Mungbean seedcoat were screened by the separated population grouping method(BSA).The genotype of the F2 population was identified,and the color genes of the Mungbean seedcoat were preliminarily mapped using Joinmap 4.0 software.The main results are as follows:1.The seedcoat color of Mungbean is controlled by 2 alleles,and there is dominant epistasis between genes.The results showed that the F2 generation of combination 1(black Mungbean×Green Mungbean)had black Mungbean:Green Mungbean =3:1,combination 2(black Mungbean×yellow Mungbean)had black Mungbean: Green Mungbean: Yellow Mungbean = 12:3:1,and combination 3(Green Mungbean × yellow Mungbean)had Green Mungbean:Yellow Mungbean = 3:1.According to the segregation ratio of offspring,it is inferred that the color of Mungbean seedcoat is controlled by 2alleles,and there is dominant epistasis.2.Two loci related to the color of Mungbean seedcoat were preliminarily located.The SSR markers were screened by the parents of two combinations of Jilu No.9× Resource 330 and BIS9805 × Resource 330.At first,83 and 60 polymorphic markers were screened out respectively,and then 3 and 2polymorphic markers were screened out by BSA.180 individual plants of F2 population were analyzed by the final selected markers,and the genetic linkage map was constructed by the software of joinmap 4.0.Sc1 located in combination with Jilu No.9 × Resource 330 was linked with 3 SSR markers in the order of Vr3-627,G3627,Xand Sc1,and the distance between Sc1 and the nearest marker X162 was 25.3c M;Sc2 located in combination with BIS9805 × Resource 330 was linked with 2 SSR markers in the order of Vr1-2463,Sc2 and X95,and the distance between Sc2 and marker Vr1-2463 was 6.9cM,and the distance from marker X95 was 19.0cM.
