Wheat stripe rust(pathogen: Puccinia striiformis f.sp.Tritici,Pst)is devastating to wheat production.It is of great significance for the sustainable development of agricultural production to discover new sources of resistance and broaden resistant genetic resources.The research includes two aspects,namely,identification of adult resistance to stripe rust of 400 wheat varieties(lines)and location of a resistance gene in wheat cultivar Xinong 529.1?Identification of adult-stage resistance to stripe rust from 400 wheat varieties(lines).In order to identify the stripe rust performance and distribution of resistance to stripe rust genes in 400 wheat cultivars(lines)from the main areas of Southwest Winter Wheat area and the northern winter wheat area,we are investigated of the adult-plant resistance with mixed races CYR31,CYR32 and CYR33 in field.Meanwhile,7 molecular markers for the known resistance genes Yr5,Yr9,Yr10,Yr15,Yr17,Yr18 and Yr26 were used to detect the resistance genes.Two aspects of data were used to comprehensively evaluate the disease resistance of the test materials.The results showed that among the 400 test materials,there were 177 varieties(lines)with high resistance to immunization(IT = 0-1)in the adult stage,accounting for 44.25%.62 intermediates(IT = 2)varieties(lines),accounting for 15.5%;medium or high-sensitivity(IT = 3/4)varieties(lines)162,accounting for 40.25%.Combined with disease resistance and known Yr genes Molecular detection analysis showed that 121,96,10,19,150,16,and 129 cultivars(lines)containing Yr5,Yr9,Yr10,Yr15,Yr17,Yr18 and Yr26,accounting for 30.25%,24%,2.5%,4.75%,37.5%,4% and 31.25%,respectively.118 materials carried two genes,30 materials carried three genes,Line CD0151270-1 carries four genes: Yr9,Yr15,Yr17 and Yr26.In addition,there are 40 materials that are not found to carry these 7 genes,and Among them,25 materials were estimated to carry unknown or new genes for resistance to stripe rust based on the disease resistance phenotype data at the adult stage.The results of this study established a wheat stripe rust resistance source identification and evaluation system,and screened 177 resistant materials with different disease resistance characteristics,of which 25 may contain the new source of resistance has laid the foundation for further cultivation of new varieties resistant to stripe rust.2?Location of a resistance gene for puccinia striiformis f.sp.tritici in wheat cultivar Xinong 529.Xinong 529 is a high-quality,strong-gluten wheat variety,which is particularly outstanding in its ability to resist stripe rust compared to other varieties,but its disease resistance mechanism is not yet clear.We crossed Xinnong 529 with the susceptible parent Huixianhong to construct a F2 mapping group with 220 individual plants and used the stripe rust race CYR33 for seedling resistance Identification and genetic analysis.Among the identification results,166 plants were resistant(IT = 0 ~ 2)and 54 plants were susceptible(IT = 3 ~ 4).After chi-square fitness test,Xinong 529 is a dominant resistance to stripe rust gene carrying a high resistance to CYR33,tentatively named Yr XN529.Using 576 SSR markers to screen the parents for polymorphism,combined with the analysis of resistance pool,finally found 4 SSR markers linked to this gene,namely Xcfd48,Xwmc134,Xgpw5195.And Xgpw7296,and use the Chinese spring-deficient tetrasomal mapping transformation gene to locate in the 1B chromosome region.The resistance spectrum identification of Xinong 529 revealed that it was a single gene line with Yr10,Yr15,Yr24,Yr26,Yr29,and Yr CH42,or that Xinong 529 genealogy analysis and resistance to stripe rust were performed.Possibly from Xiaoyan 597,a male parent with a genetic background of Changsuiyan wheatgrass.This is different from the source of genes on many 1B chromosomes.Therefore,the gene for resistance to stripe rust Yr XN529 may be a new gene with further research value.
