Danshen is the dried roots and rhizomes of Salvia miltiorrhiza Bge.,which belongs to the Salvia genus of Lamiaceae family.It is one of the most important traditional medicinal materials in China.It has the functions of promoting blood circulation and removing blood stasis and menstrual pain relief.It is mainly and clinically used for various cardiovascular and cerebrovascular diseases.Recent studies have shown that the medicinal plant Salvia miltiorrhiza has a diverse community of endophytic fungi,part of which have been proven to not only effectively promote plant growth and bioactive compounds accumulation,but also produce secondary metabolites with rich and chemical diversity.Chaetomium globosum D38 is a functional endophyte isolated from the roots of S.miltiorrhiza.Previous studies have shown that D38 elicitor can significantly increase the content of tanshinones in the hairy roots of S.miltiorrhiza,and C.globosum D38 can promote both the growth of S.miltiorrhiza.and the accumulation of active ingredients.In order to explore the potential biologically active compounds in the secondary metabolites of C.globosum D38,we conducted solid fermentation of C.globosum D38 in rice medium and subsequently employed a variety of column chromatography methods,including silica gel,Sephadex LH-20and ODS,for compounds separation and purification.The structures of isolated compounds were further identified using 1D and 2D NMR,mass spectrometry,single crystal diffraction and other spectroscopy techniques,combined with literature data comparison.As a results,1)A total of 9 compounds were isolated and identified,including two new compounds,among which,there were five complex indole alkaloids:two new compounds:salchaetoglobosin A?3?and salchaetoglobosin B?10?,and three related know analogues:cheatoglobosin E?1?,chaetoglobosin Fex?5?,chaetoglobosin Vb?6?,and 5 other known small molecule compounds:n-tritriaconta-4,12-diene?2?,?9Z,12Z?-nonadeca-9,12-dienoic acid?4?,p-hydroxybenzoic acid?8?and 2,2'-dihydroxyl ether?9?;2)The CCK-8 method was used to evaluate the proliferation inhibitory activities of the new compound salchaetoglobosin B?10?on colon cancer cell HCT-116and prostate cancer cell PC-3.The results showed that salchaetoglobosin B has moderate inhibitory effects on both HCT-116 and PC-3 cells,with IC500 values of 60.42?M and 62.21?M,respectively.This study preliminarily clarified the secondary metabolites of the endophytic fungus C.globosum D38 of S.miltiorrhiza,which provided a basis for further research on the material basis of this functional fungus in facilitating the growth of S.miltiorrhiza and promoting the accumulation of bioactive compounds.
