Aconitum barbatum Pers.var.hispidum?DC.?Seringe and Aconitum flavum Hand.-Mazz are perennial herbaceous of Ranunculaceae aconitum,both roots can be used as medicine,it is a typical famous medicine,and it is the main raw material for aconitine extraction.Nowadays,the demand for aconitine in the field of biomedicine development is increasing,and the wild Aconitum barbatum Pers.var.Hispidum?DC.?Seringe and Aconitum flavum Hand.-Mazz seed reproduction rate is very low,and the wild resources are over-extracted,resulting in annual reduction in reserves cannot meet market demand.Then artificial introduction,domestication,cultivation and reproduction are the only ways to solve the lack of resources.However,in the actual production process,there are two main dilemmas:First,there are obvious dormancy phenomena in the seeds,which causes great troubles for sowing.Second,the low temperature affects the yield and quality of medicinal materials.Therefore,this article aims to explore the dormancy factors of the seeds of these two plants and how to break dormancy;moreover,to improve the cold resistance of the seedlings of the two plants provides a cold-proof technical theory for the cultivation of medicinal materials.The seeds of Aconitum barbatum Pers.var.Hispidum?DC.?Seringe andAconitum flavum Hand.-Mazz were used as experimental materials.The seeds viability were measured by TTC staining method,and the water absorption of the seeds were measured.The crude body fluid inhibitory activity was tested with wheat and cabbage seeds and its own seeds.Method to explore the factors of the seeds dormancy;Use different types and concentrations of chemical agents to explore the best method to break the seeds dormancy;find the optimal temperature for the seeds germination through different temperature gradients;a light incubator was used to simulate the low temperature environment,and the effect of spraying SNP solution on the cold resistance of Aconitum barbatum Pers.var.Hispidum?DC.?Seringe and Aconitum flavum Hand.-Mazz seedlings were investigated.The research results are as follows:1.The seeds have good water absorption,the seed coat does not have mechanical resistance,and it will not cause water absorption obstacles,so it is not a cause of dormancy.2.The seed viability of the Aconitum barbatum Pers.var.hispidum?DC.?Seringe and Aconitum flavum Hand.-Mazz is very high,and the seed embryo is mature,which is not a factor that causes the seeds germination rate to be low.3.Crude extract of the seeds have obvious inhibitory effects on the germination and seedling growth of other plant seeds and its own seeds,and the greater the concentration of the crude extract,the stronger the inhibitory activity,indicating that the inhibitory substance contained in the crude extract can inhibit the germination of other plant seeds and their own seeds,so the presence of endogenous inhibitory substances plays a leading role in the factors of seed dormancy.4.Compared with static water soaking,running water rinsing significantly increased the germination rate of seeds,indicating that running water rinsing can reduce?or dilute?the endogenous inhibitory substances of the seeds,further indicating that the dormant factor of the seeds is the presence of endogenous inhibitory substances.5.The effects of different temperatures on the seeds germination of Aconitum barbatum Pers.var.hispidum?DC.?Seringe and Aconitum flavum Hand.-Mazz were investigated.The results showed that:The optimal temperature for seed germination of Aconitum barbatum Pers.var.hispidum?DC.?Seringe at 20?;the optimal temperature for seed germination of Aconitum flavum Hand.-Mazz at 16?.6.GA3,KH2PO4,KMnO4,H2O2 can break the dormancy of the seeds,improve the germination rate,germination potential,vitality index of Aconitum barbatum Pers.var.hispidum?DC.?Seringe and Aconitum flavum Hand.-Mazz,and shorten the germination time lag.In particular,500 mg/L GA3 had the best treatment effect on Aconitum barbatum Pers.var.hispidum?DC.?Seringe seeds,germination rate increased from 44%in natural state to 76%;250 mg/LGA3 had the best treatment effect on the seed of the Aconitum flavum Hand.-Mazz,germination rate increased from 16.56%in natural state to 85.00%.7.The optimal concentration of 200?mol/LSNP solution to Aconitum barbatum Pers.var.hispidum?DC.?Seringe seedlings and 150?mol/L SNP solution to Aconitum flavum Hand.-Mazz seedlings seedlings significantly increased SOD,POD,CAT activity,soluble sugar content,proline content,chlorophyll?a+b?content.reduces the accumulation of malondialdehyde and hydrogen peroxide harmful substances.It is shown that the seedlings of Aconitum barbatum Pers.var.hispidum?DC.?Seringe and Aconitum flavum Hand.-Mazz show resistance to cold environment.
