A large number of China's unique cryptomeria fortunei ancient trees are distributed in Zhejiang province in the west Tianmu mountain national nature reserve,there were more than 2000 cryptomeria fortunei over 50 cm in diameter among them.There are also a lot of trees that are thousands of years old in Zhejiang province in the west Tianmu mountain national nature reserve,whose most trees are over 100 years old,which is the largest collection of ancient C.fortunei trees in the world.The C.fortunei trees are distributed like a forest from Zen source temple at the foot of the mountain to the top of the old hall.However,there has been a serious case of decay and death,a large number of ancient C.fortunei trees have died,population decline intensified in recent decades.This experiment focuses on the whole picture of Tianmu mountain,investigating and analyzing the decay of C.fortunei trees in Tianmu mountain,the cause of the rot of C.fortunei and community dynamics of fungi and bacteria in C.fortunei trees with different decay degrees under natural conditions,based on which we use an antagonistic bacillus was isolated investigate the preliminary control effect of Bacillus on potential rot fungi.The results showed that the occurrence of ancient C.fortunei trees putrefaction in western Tianmu mountain was positively correlated with the road direction and crown width of the stumpers,the degree of decay was divided into 4 grades according to the ratio of area,depth and diameter.This paper analyzes and statistics the microbial groups,related enzymes and cellulose and lignin content indexes in the disease sites with different grades of decay and preliminarily establishes the disease indexes of grade 1-4.According to the severity of disease index,the microbial diversity index was 0.9,0.9,1.0 and 1.0.Fungal index?×106 cfu/g?: 25,610,1115 and above;cellulase activity index?U/g.48 h?:0.20.3,0.30.5,0.50.7 and above;xylanase activity index?U/g.120 h?:0.10.2,0.20.4,0.40.6 and above;pectinase activity index?U/g.120 h?:0.2,0.3,0.4 and above;cellulose content index?%?:0.420.49,0.320.41,0.270.31 and below.In addition,it was found that compared with the bacterial community,the most notable changes are the three genera Parcubacteria,nv.OPS17,Aquincola,while the fungi?Coniophoraceae,Simplicillium,Trechisporales,Trechispora,Penicillium,Lecanoromycetes,Capnodiales?are 7 genera.the fungal community was more sensitive to the change of decay degree through high-throughput analysis of the fungal and bacterial communities of ancient Cryptomeria fortunei trees with different decay degrees in Tianmu mountain under natural conditions.The analysis between intergroup differences between dominant species bacteria and fungi in different decay degrees showed that there was no difference in bacterial composition at different levels of decay.There are significant differences in fungi.The VEEN graphs results showed that the most endemic fungi were found in the ancient C.fortunei trees with the lowest degree of decay,the fungus characteristic OTU decreased in the moderate decay degree and those with the most decay had the most endemic bacteria.The interaction between the fungus and bacteria community of ancient cryptomeria fortunei with lignin and cellulose environmental factors was analyzed.There was a significant decrease in cellulose content and there was a significant positive correlation between 5 fungal OTU and cellulose content with the lignin content did not change significantly with the increase of the degree of decay.There was no significant correlation with bacterial OTU.The community network map shows that actinomycetes may be an important participant in the decay process.Six strains of fungi with the ability to degrade cellulose screened by cellulose Congo red medium were thought to potentially deepen the decay of C.fortunei.It was found that neither chemical reagent?Azoxystrobin,Difenoconazole,Tebuconazole,Carbendazim?nor biological control alone could inhibit all six strains of fungi inhibited by chemical bacteriostatic agents and bacillus methylotrophic?L-01?.Moreover,it is difficult to show a good inhibitory effect on all isolated strains by chemical reagent or biological control alone,the inhibitory rates of the recommended concentrations on diseased microorganisms were only 41.8%,47.6%,45.3%,and 50.1%.The average inhibitor of L-01 is 49.3%,which is equivalent to the commonly used chemical reagents.The results showed that which has laid a foundation for disease detection,decay prevention and treatment of ancient cryptomeria fortunei trees in Tianmu mountain and the prevention and treatment of ancient C.fortunei protection and decay.
