Animal acariasis,a popular worldwide parasitic disease,caused reducing the productivity and quality of animal products,increasing the cost for feeding and treatment.It caused great economic losses and even endangered human health.Previous research revealed,the Eugenia caryophyllata extract microemulsion had no irritation to rabbit skin and conjunctiva,coincide with the demands for dermal administration preparation.The Eugenia caryophyllata extract microemulsion had significant acaricidal activity to Psoroptes cuniculi in vitro.According to the Technical Guiding Principle for Veterinary Drug Researches,the present study performed both experimental clinical trial and expanded clinical trials for Eugenia caryophyllata extract microemulsion against Psoroptes cuniculi,the following main results were obtained:Experimental clinical trials:Fifty rabbits,naturally infected with Psoroptes cuniculi and met standard requirements,were randomly divided into 5 groups,10 for each group.The 5groups were high-,middle-and low-dose treated groups?embrocated 4,2,or 1 times per day;3 days?,ivermectin drug control group and disease control group.In addition,10 healthy rabbits were selected as healthy control group.On the 4thday,hematological indexes were detected.On the 7thand 14thday,the therapeutic effects were evaluated.By the end of the experiment,histopathological examination was performed to experimental rabbits.The results showed,after treatment with Eugenia caryophyllata extract microemulsion,the ear symptoms were improved,the scabs fallen,and the number of mites reduced significantly.The hyperkeratosis,acanthosis and derma cell inflammation were relieved.The symptom scores and the mites decrement rate of high-and middle-dose group were significantly different from low-dose group?P<0.01?,the effective rates and mite clearance rates were all100%.WBC,Lym,Gra,RBC and HGB in each group were no significant difference with drug control and disease control groups?P>0.05?.ALT,AST,TP,ALB,CK and CRE in each group were no significant difference with disease control and healthy control groups?P>0.05?.ALB in drug treated groups were significantly lower than healthy control group?P<0.01?.AST of high-dose group was significantly lower than healthy control group?P<0.01?.there is no regular pattern in BUN change.No abnormal changes was observed by dissecting,heart,liver,spleen,lung,kidney,stomach and intestine indexes of each dose treated group were without significant difference with disease control group?P>0.05?.The heart indexes in different dose treated groups and stomach index in high-dose group were significantly higher than healthy control group?P<0.01?.No significant abnormality of histopathological was observed in the high-dose group.The results indicated that Eugenia caryophyllata extract microemulsion had remarkable therapeutic effects on Psoroptes cuniculi without side effects.Direction for use of Eugenia caryophyllata extract microemulsion was:embrocate 2times per day,continue for 3 days.Expanded clinical trials:Sixty Psoroptes cuniculi naturally infected rabbits were randomly divided into 2 groups,30 for each group.The two groups were Eugenia caryophyllata extract microemulsion group?embrocated Eugenia caryophyllata extract microemulsion 2 times per day,3 days?and ivermectin drug control group.The results showed that,on the 14thday after treatment,the clinical symptoms were significantly relieved.The symptom scores were reduced from 3.27±0.77 to 0.51±0.71,effective rate was100%,mite clearance rate was 93.3%.The therapeutic effect was equal to ivermectin solution?P>0.05?,and no abnormal appearance was observed.The above results indicated that,according to Eugenia caryophyllata extract microemulsion clinical direction,the Eugenia caryophyllata extract microemulsion had promising acaricidal activity against Psoroptes cuniculi,without side effects.
