Tomato(Solanum lycopersicum)is one of the important economic benefits of vegetable crops,and its economic benefits are affected by flower and fruit falling.IDA encodes a small peptide,which sends signals(HAE)and HAE-LIKE2(HSL2)through leucine rich repeat receptor like kinase haesa to control floral organ abscission.It is found that IDA-HAE-HSL2 pathway in Arabidopsis may be independent of ethylene mode to control abscission,while it is not clear how IDA-HAE-HSL2 regulates abscission in eggplant.In this paper,we use biological analysis to screen and study the IDA1,IDA7 and IDA8 that regulate abscission.The purpose of this study was to reveal the regulatory role of IDA gene in tomato flower stalk,and the regulatory pathway and mechanism related to ethylene.The study on the mechanism of fruit abscission has certain guiding significance to the regulation of fruit development and cultivation management in production.The main results are as follows: 1.The gene family scan of 11 tomato IDA family members was excavated.The real-time quantitative results showed that the tomato IDA1,IDA7,and IDA8 were specifically expressed during tomato abcission.Treatment of tomato flower stalks with in vitro synthetic peptides showed that when the concentrations of SlIDA1,SlIDA7,and SlIDA8 peptides were 0.01?M,there was no effect of inducing abcission,and at 1?M,it was possible to promote flower stalk abcission.When the concentration was 10?M,it significantly promoted flower stalk Abcission,but the effect of 100?M induced abcission was not different from 10?M.2.SlIDA1,SlIDA7,and SlIDA8 gene knockout plants were constructed using CRISPR / Cas9 technology and their abcission rates were determined.The results showed that CR-slida1,CR-slida7,and CR-slida8 delay abcission.3.Ethylene treatment can promote the abcission process of CR-slida1,CR-slida7 and CRslida8,but its promoting effect is smaller than that of the control.Application of SlIDA1,SlIDA7 and SlIDA8 peptides could significantly accelerate 1-MCP,indicating that IDA acts on downstream regulation of abcission of ethylene.4.The expressions of cellulase and pectinase were determined on the knockout plants.The results of real-time quantitative PCR showed that the expressions of cellulase and pectinase in the knockout plants CR-slida1,CR-slida7 and CR-slida8 They are all adjusted to varying degrees.
