E.coli diarrhea in piglets is one of the most common infectious diseases with extremely high morbidity and mortality in modern animal husbandry.Inflammation of the intestine widely affects the development of animal husbandry.At present,antibiotics and cortisol drugs are mostly used to treat these two diseases.Although they have certain effects,they have great side effects and are easy to cause antibiotic abuse and lead to microbial resistance.Under the trend of "replacement of antibiotics" in animal husbandry,finding an effective antibacterial and anti-inflammatory product has become the primary task.Sodium humate is a macromolecular organic substance rich in organic matter and a large number of hydroxyl and carboxyl groups.It has anti-inflammatory,antibacterial,anti-ulcer,astringent,antidiarrheal and other effects.This experiment aims to use dextran sulfate sodium(DSS)to successfully establish a model of ulcerative colitis,and use E.coli K88 and DSS to establish a piglet E.coli diarrhea model to study the effects of sodium humate on these two intestinal diseases.In response to the above content,this experiment carried out the following research work:(1)Using plate colony counting and scanning electron microscopy,the effects of sodium humate on Escherichia coli K88,Staphylococcus aureus,and Salmonella were studied.It is concluded that sodium humate has a significant inhibitory effect on Escherichia coli and Staphylococcus aureus,and the inhibition rate of 0.4% sodium humate on Escherichia coli reaches 77.27%.Scanning electron microscopy shows that sodium humate can cause the surface of E.coli to collapse,fold,and overflow the contents.The inhibition rate of 0.4% sodium humate on Staphylococcus aureus reached 13.68%.After adding sodium humate,the colonies of Staphylococcus aureus were obviously small,the shape was wrinkled,and the shape changed.(2)Ulcerative colitis model was successfully established by using 3.5% DSS,and it was concluded that 0.4% sodium humate can repair intestinal villi,inhibit inflammatory cells,and regulate the balance of intestinal flora,indicating that 0.4% sodium humate can relieve DSS-induced Inflammation of the intestine.(3)The mice diarrhea model was successfully induced by E.coli K88 and DSS.The duodenal inflammation of the model mice was the most serious.The levels of LPS,TNF-?,IL-1? and IL-6 in the blood increased significantly,the intestinal flora was disturbed,and the abundance of Proteobacteria increased.After modeling,mice ingested 0.4% sodiumhumate,the LPS,TNF-?,IL-1? and IL-6 concentration decreased,the balance of intestinal flora was adjusted,the intestinal tissue was repaired,and the symptoms of enteritis were significantly relieved.It shows that 0.4% sodium humate can effectively alleviate the intestinal inflammation induced by the combination of E.coli K88 and DSS.
