Embryo transfer is a very important procedure in embryo engineering,which plays an important role in livestock breeding.In this study,mice were used as the model to study the effect of embryo secretion on the efficiency of embryo transfer.The mouse fertilized embryos were cultured in vitro,and embryo culture medium was collected,and exosomes secreted by the embryos were extracted by ultracentrifugation.The exosomes was round or oval cup-shaped,which has a typical characteristics of exosomes.Mi RNA was extracted,and followed RT-q PCR.Mir-25,mir-302 c and mir-191 were detected in the culture medium.Bioinformatics results suggested that mi RNA in the culture medium played an important role in the development of embryo.Further followed biological experiments verification,the results showed that there was no significant difference in embryo transfer efficiency between the transfer medium with or without the secretion groups.It is speculated that the amount of embryo transfer fluid is only a small part of the amount of culture fluid,and it does not completely contain all embryo secretion,so it may be related to the lack of secretion;on the other hand,the composition of embryo secretion is not clear,there are some components beneficial to embryo,but it is also unable to exclude the components harmful to the development of embryo.Therefore,in the follow-up study,it will be of great significance to improve the efficiency of non-surgical embryo transfer by controlling the single factor variables of the secretion derived from embryo cultured in vitro.To sum up,we found that exosomes and mi RNAs are secreted by embryos in vitro.These secretions may play an important role in the development of embryos which is of great significance to improve the efficiency of embryo transfer.
