| Non-surgical embryo transfer in mice has a lower success rate than surgical embryotransfer, and this limits the wide application of the non-surgical embryo transfer. Comparedwith surgical embryo transfer, non-surgical embryo transfer was simpler, lower pollution,lower cost, and smaller injury for mice, also without professional surgical instruments. Ourmethod was as efficient as surgical embryo transfer however it was much simpler, easierand more significantly eliminates surgical trauma to the recipient. The experiment from theanesthetic type, hormone regulation, false pregnant rat days, material implantable devices,the graft diameter, tube length, curvature, simulated transplantation, clocked liquid to thevolume of air, moved aspects embryo number and transplantation method has beendeveloped, traditional non-surgical embryo transfer methods has been improved, thesuccess rate of embryo transfer has been increased.In anesthetic experiments, the results showed that the differences between theanesthetic and the control group were not significant (P>0.05); The results show that theefficiency of2.5d and3.5d pseudopregnant mice transplant surgery method wassignificantly different (P<0.05), the efficiency of non-surgical transplantation differencewas not significant (P>0.05), and efficiency of2.5d pseudocyesis rat surgical andnon-surgical method transplantation was significantly different (P<0.05), efficiency of3.5dpseudopregnant mice non-surgical method was not significantly different.After embryo transfer, the two group mice were injected0.1ml respectively20ug/mlof estrogen and progesterone through the subcutaneous injection, efficiency of addingprogesterone after transplantation was only35.7%; In the supplement estrogen mice onlyone was pregnant, others were failed.Using glass tube and the transplantation of PTFE tube, the farrowing rate were55.9%and72.4%respectively, significant difference (P<0.05); Also using PTFE tube, differentdiameter (450μm,600μm,750μm), the impact on the success rate of transplantation isdifferent also, farrowing rate of transplant tube with Group600μm and Group750μm was72.7%and54.2%, significant difference (P<0.05), Compared with other two groups, Seedrate of Group450μm was70.5%, no significant difference; Four groups of transplant tubelength with1.5cm,2.0cm,2.5cm and3.0cm, seed rates were60.4%,64.0%,71.1%and 60.4%respectively, no significant difference (P>0.05).1-cell embryo,2-cell embryo in vitro culture to the blastocyst stage (E3.5d) weretransplanted in2.5d pseudopregnancy mice, IVF embryos cultured to blastocyst stage weretransplanted in3.5d pseudopregnancy mice. Results showed that compared the surgical andnon-surgical methods, no significant difference (P>0.05).The traditional non-surgical embryo transfer was improved, success rate oftransplantation was increased, the efficiency of non-surgical embryo transfer was the samewith surgical embryo transfer. And surgical embryo transfer was complied with theprinciple of experimental animal ethics. Importantly, the research model was provided forthe embryo transfer of human, cattle and other large animals. |
