Manshanhong which is also named Yingshanhong,derived from the dried leaves of Rhododendron dauricum L.,with the function of relieving cough and reducing sputum,it is commonly used to cure the acute and chronic bronchitis in clinic.This plant mainly distributes in the northeast of China,for example Heilongjiang?Jilin?Liaoning and Neimenggu Province,etc.As a common raw material for various Chinese patent medicine,Manshanhong extract powder is the water decoction after removing oils.In order to develop and utilize this medicine well,we need to find the important bioactive components and comprehensivly control the quality.In the study,flavonoids were seperated and purified,at the same time,the fingerprint of Manshanhong extract powder was established.All of these could control the quality of Manshanhong extract better.Contents:1.The research focus on and seperation and identification of chemical components.Solvent method and a variety of modern chromatographic separation techniques-(including polyamide column,ODS,Sephdex LH20 and HPLC ect)were used-.At last,five chemical constituents were isolated.-On the basis of measurement of physico-chemical characters,NMR data analysis and chemical method,the structures of five compounds were respectively identified as q uerce tin(compound I)?3'-OH-farrerol(compound II)?Poriol(compound III).Farrerol(compound IV)?Hyperoside(compound V).This work laid a consistence foundation for futher understanding the composition of Manshanhong extract powder.2.Fingerprint was preliminary established and flavonoids,the main active components were further studied.HPLC-DAD method was used to analyze the purity of main marker peaks.Combined with the reference substance,8 character peaks were identified and seperated completed.Furthermore,the contract study was carried on between medicinal materials and extract,more comprehensive relevances were found,which has practical significance for its quality control.3.We have a study on analysis method in the quality of extract powder.For example,a thin layer chromatographic identification method of Manshanhong extract powder was eatablished.And multi-componets assay was built up by HPLC,in this method,double wavelength switching method was used to determine the content of Hyperoside,quercetin and Farrerol.Through the methodology validation,the precision,stability,repeatability and recovery all meet the requirements.Purposes:To further comprehensive elaboration the efficacy of the extract,furthermore,establish and perfect the analysis method of flavonoids in extract,the quality could be Better control.
