| [Objective]To explore a more stable model of ankle joint urate arthritis in rats.To observe the effects of pricking blood therapy on acute gouty arthrits rats model’s local symptoms,morphological changes under light microscope and ultrastructure under electron microscope,and explore the effector mechanism of pricking blood therapy’s treatment.[Method]Forty SD rats were equally divided into normal control group,model group,drug group and pricking blood group randomly.By injecting uric acid into the right ankle joint cavity so as to set up the acute gouty arthrits model.The drug group received 120 mg/kg gastric perfusion of ibuprofen after 6h and 24h;the pricking blood group pricked the right Kunlun points after 6h and 24h.We Measured and recorded the cross section diameter of the injected right ankle joint of the forty rats before modeling,and also the numbers after 6h,24h,48h(before dead)when modelized;and to observe the morphological changes under light microscope and ultrastructure under electron microscope;and used elisa to detect the content of IL-4 and IL-10.[Result]According to the result of light microscope,pink crumby substance deposition has been found in model group with dense inflammation infiltration;there were some pink crumby substance deposition and inflammation infiltration in drug group;small mount of pink crumby substance deposition and inflammation infiltration were in pricking blood group.As electron microscope showed,about model group,a few of articular cartilage cell membrane was damaged.Exudate,cell process endopasmic reticulum expansion have turned up.Arrangement of collagen fibers was disorder and dense;Exudate;cell process and endopasmic reticulum expansion have been found in drug group.Mitochondria decressed and Arrangement of collagen fibers was in disorder;there was a few exudate in articular chondrocyte of pricking blood group.Basic cell structure was normal and arrangement of collagen fibers was disorder to some extend.The difference of IL-4’s content between normal group and other 3 group was statistically significant(P<0.05).Drug group,pricking blood group and model group was statistically significant(P<0.05).Pricking blood group and drug group was no statistically significant(P>0.05).The difference of IL-10’s content between model group and normal group was statistically significant(P<0.05).The difference of drug group,pricking blood group and normal group was no statistically significant(P<0.05).The difference of drug group.pricking blood group and model group was statistically significant(P<0.05).The difference of pricking blood group and drug group was statistically significant(P<0.05).[Conclusion]Ibuprofen dosing and pricking blood therapy for acute gouty arthritis have good therapeutic effect.It’s rapider decrease of a swelling by pricking blood therapy than Ibuprofen dosing.Pricking blood therapy can reduce the deposition of uric acid sodium in articular cavity,lessen exudate,and improve the ulrtastructure of arthrodial cartilage quick and effectively and compared with conventional therapy,pricking blood therapy is better effective,faster and less side-effect.But from the point of view of anti-inflammatory factor(IL-4,IL-10),it’s better by Ibuprofen dosing than pricking blood therapy.Pricking blood therapy is by adjusting the content of local anti-inflammatory factor to achieve anti-inflammatory effect. |
