| Traditional Chinese herb YZ,as the folk medicine,has been widely used for 20 years,in the southwest of our country among the ethnic minority areas.After investigation,the drug is good for function of anti-hemostasis,anti-inflammatory analgesic and enhancing immunity,which has definite clinical effect.For the reason that a lot of folk medicine were not included in the pharmacopoeia in our country and overseas,there was no relevant research reports about the date of YZ.This study on the anti-inflammatory effects and mechanism of YZ contains the following three sections.1.Preparation and separation of the extract of Chinese traditional medicine YZTo establish the extraction methods of traditional Chinese medicine YZ,and calculate the extraction efficiency,Chinese traditional medicine YZ were prepared through being crushed,sieved and dry.The extract and separation of traditional Chinese medicine YZ were received though Solvent extraction method and extraction separation process.The results showed that alcohol extract of traditional Chinese medicine YZ and five different parts were received though the solvent extraction method and extraction separation method,respectively were petroleum ether part,chloroform part,ethyl acetate parts,n-butanol parts and water phase parts.This indicated that weight of Chinese medicine YZ alcohol extract was 114.4 g,extraction ratio was 22.8%.Parts of the petroleum ether extract 9.1 g,1.8%;Chloroform part extract 5.1 g,1.7%;Ethyl acetate parts extract 11.2 g,3.7%;Normal butanol part extract weight 44.0 g,8.8%;Parts of water extract weight 26.0 g,5.2%.2.Anti-inflammatory effects of the different parts of Chinese medicine YZ alcohol extractTo find out the best parts that can effectively restrain the role of inflammation of different parts of accohol extraction of Chinese tradition medicine YZ through the animal inflammation model,70 male kunming mices were used for the experiment,compare mice auricle swelling degree and swelling inhibition rate with the experimental group and control group through the mouse xylene-induced auricle swelling model.80 SD rats were used in the experiment of the modle of the Angle fork lai glue which can make the rats foot swell,the rats foot swelling degree with volume method were compared between each treatment group and control group after the treatment in 0～6 h.70 male kunming mices were used for glacial acetic acid that can increase capillary permeability of mice,test absorbance of mice abdominal cavity effusion in 590 nm in order to reflect capillary permeability.The results showed that the mice ear swelling degree of chloroform part of traditional Chinese medicine YZ alcohol extract was significantly different(P<0.05)with physiological saline group;Compared with saline group,aspirin mice ear swelling inhibition rate was 45.03%,Chinese traditional medicine YZ alcohol extract chloroform part 41.81%,the difference was not significant(P>0.05).After being injected carrageen in 2～6 h,rats foot swelling degree of chloroform part of Chinese traditional medicine YZ alcohol extract group was significantly different with physiological saline group,but did not show significant difference with aspirin group and glycyrrhizic acid diamine group.Chloroform part group and the water phase parts mice celiac collecting liquid absorbance and physiological saline group significant difference(P<0.05).This indicated that the traditional Chinese medicine YZ alcohol extract chloroform part had a significant anti-inflammatory effect though out the three experiments,and the water phase site groups also had certain anti-inflammatory effects.3.Initial study on mechanism of anti-inflammatory action of YZTo detect the inflammation factors including PGE2,MDA,5-HT and NO,preliminary explore the anti-inflammatory mechanism of Chinese medicine YZ extract chloroform parts,120 SD rats were used with carrageenin to induced rats toes swelling,detection the PGE2,MDA and 5-HT content of rats foot inflammation tissue.40 kunming mices were used with the glacial acetic acid to induced increase of capillary permeability in mice,detecting the NO content of mice serum.The results shows that PGE2 and 5-HT content of traditional Chinese medicine YZ alcohol extract chloroform part group of big rat toes swelling were significant different with physiological saline group(P<0.05),but not with aspirin group and glycyrrhizic acid diamine group(P>0.05).In contrary,the MDA content of Traditional Chinese medicine YZ alcohol extract chloroform part group of big rat toes swelling of the parts of the tissue was not significantly different with normal saline groups(P>0.05),but significant different with aspirin group and glycyrrhizic acid diamine group(P<0.05).NO content of Traditional Chinese medicine YZ alcohol extract chloroform was significantly different with physiological saline group(P<0.05),but not different with aspirin group and glycyrrhizic acid diamine group(P>0.05).This indicated that Chinese medicine YZ extract chloroform part could obviously reduce PGE2,5-HT,NO content in inflammation tissue,it certainly had similar anti-inflammatory effect as aspirin and glycyrrhizic acid diamine group,but did not related with MDA content. |
