Dictyophora indusiata is one of good-sized precious edible and medicinal fungus in the world,and it is also a kind of rare specialty in China.The sporocarp of Dictyophora indusiata is tasty and full of nutriments,and beyond that,it has certain medicinal value.This subject takes Dictyophora indusiata as material,which was produced by Fujian Changshun,and studying on isolation,purification and antioxidant activity of Dictyophora indusiata polysaccharide.The result can provide a basis of the further development and application of Dictyophora indusiata.(1)To analyze the basic element of sporocarp with modern technical methods and to test the contents of amino acid,protein,crude fiber,water-soluble polysaccharides and trace elements,the result showed that Dictyophora indusiata contains crude polysaccharide,protein and moderate crude fibre,and it is low in fat.It was a kind of good food in low fat and high protein,with dietary fiber.(2)Using hot water extraction,taking extraction temperature,extraction time and solid-liquid ratio as the indexes of Dictyhora indusiata polysaccharide's extraction rate and by means of single factor experiment and orthogonal experiment,the optimum extraction process of Dictyophora indusiata polysaccharide which was extracted by hot water has reached.Optimum extraction conditions:solid-liquid ratio 1:50,extraction temperature of 95 ?,and extract time 2.5h.The extraction rate of Dictyophora indusiata polysaccharide is 16.8%under the optimal conditions.(3)Polysaccharide fractions were classified by sephadex column chromatography initial.The extractions which were from the polysaccharide of Dictyophora industiata sporocarp can obtain four constituents of Dictyophora indusiata polysaccharide by DEAE Sephadex A-50 chromatography.They are named GLPS-1?GLPS-2?GLPS-3 and GLPS-4 respectively.Evenly distributions of GLPS-1?GLPS-2?GLPS-3 and GLPS-4 were identified by Sephadex G-200.Elution curve shows that GLPS-1?GLPS-2 and GLPS-4 have the character of uniform distribution,nevertheless,the constituent of polysaccharide in GLPS-3 is more complex.(4)Antioxidant capacity of Dictyophora indusiata Polysaccharide is measured,and compared with antioxidants of astaxanthin and ascorbic acid.Taking hydroxyl radical,reducing power and DPPH free radical as study subjects,and evaluating the antioxidant capacity of Dictyophora indusiata Polysaccharide in vitro objectively and roundly.In three antioxidative evaluation systems of hydroxyl radical,reducing power and DPPH,Dictyophora indusiata Polysaccharide reflects better scavenging activity on free radical,and the antioxidant capacity is stronger as the concentration increased.In addition,they presented a good dose-effect relationship.The antioxidant capacity of Dictyophora indusiata Polysaccharide is higher than Polysaccharide GLPS-1 and the compound of same concentrations of Polysaccharide GLPS-2,Polysaccharide GLPS-4 and three kinds of Polysaccharide.In the antioxidative evaluation systems of hydroxyl radical,reducing power and DPPH,antioxidant capacity of astaxanthin is stronger than ascorbic acid in same concentration,and stronger as the concentration increasing.(5)The subject had studied the effects of Dictyophora in.dusiata group,Dictyophora indusiata:astaxanthin(3:1),Dictyophora indusiatai:astaxanthin(4:1),Dictyophora indusiata:astaxanthin(5:1),Dictyophora indusiata:astaxanthin(1:3)and Dictyophora indusiata:astaxanthin(1:2)on tissue SOD activity of Drosophila tissue homogenate,total antioxidant capacity(T-AOC),andscavenging capability to hydroxyl radical.Results indicate that the SOD activity in male Drosophila of group Dictyophora indsiata:astaxanthin(5:1)reaches significant level,and the SOD activity in female Drosophila reaches very significant level.They increase by 25.73%.28.82%respectively.The T-AOC activities in male and female Drosophila of Dictyophora indusiata:astaxanthin(5:1)group all reach significant level,increasing by 35.14%and 14.29%respectively.Inhibition of OH radical capacity inside female Drosophila in Dictyophora indusiata group,Dictyophora indusiata:astaxanthin(5:1)reaches very significant level,increasing by 5.53%?9.57%respectively.Dictyophora indusiata polysaccharide can enhance the antioxidant ability of Drosophila,and it also can eliminate internal free radicals,as that it can anti-aging.(6)This subject had studied the effects of cultures of Dictyophoraindusiata group,Dictyophora indusiata:astaxanthin(3:1),Dictyophora indusiata:astaxanthin(4:1),Dictyophora indusiata:astaxanthin(5:1),Dictyophora indusiata:asXtaxanthin(1:3)and Dictyophora indusiata:astaxanthin(1:2)on half-death period,average life-span,average life-span elongation and maximum life-span.It turns out that half-death period of male Drosophila in group Dictyophora indusiata:a.staxanthin(5:1)reaches very significant level,and half-death period of female Drosophila reaches significant level.The half-death periods all are 49d.Average life-spans of male and female Drosophila in group Dictyophora indusiara:astaxanthin(5:1)all reach significant level,and the average life-span of female Drosohphila can prolong to 49,60d,and the average life-span of male Drosohphila can prolong to 47.72d.Average life-span elongations of male Drosophilas in Dictyophora indusiara group,Dictyophora indusiata:astaxanthin(3:1),Dictyophora indusiata:astaxanthin(5:1),Dictyophora indusiata:astaxanthin(1:3)and Dictyophora indusiata:astaxanthin(1:2)are 14.33%?14.14%?14.96%?11.42%?9.71%respectively,and average life-span elongations offemale Drosophilas are 11.41%?14.72%?20.71%?9.93%?12.48%.Maximum life-span of female Drosohphila in Dictyophora indusiata:astaxanthim(5:1),group reaches very significant level;maximum life-span of male Drosohphila reaches significant level;maximum life-span of female Drosohphila in group Dictyophora indusiata reaches significant level.
