In this study,three kinds of nephrotoxic cells model of streptomycin,uric acid and hydrogen peroxide in vitro were constructed.Protective effects of extract of serum of Huangkui Capsule,Congrong Yishen Granule,Sunset Abelmoschus Flower and Cistanche deserticola Ma on toxicity of renal tubular epithelial cells?RTECs?.To establish the rat model of streptomycin nephrotoxicity injury in rats to study the protective effect of Huangkui Capsule and Congrong Yishen Granule on its injury.Combined with in vitro and in vivo experiments,investigated the mechanism of protective effect on renal tubular epithelial cell injury of Huangkui Capsule and Congrong Yishen Granule of two kinds of traditional Chinese medicine and its main components Sunset Abelmoschus Flower and Cistanche deserticola Ma.To lay a theoretical foundation for clinical application and further development.In the first part,three exogenous agents were established to induce RTECs in vitro nephrotoxic cell models.Streptomycin,uric acid,and hydrogen peroxide were used to injury RTECs.Morphological changes,cell proliferation inhibition rate and cell LDH release were used as model evaluation indexes to judge the nephrotoxicity of three exogenous substances,and the apoptosis was observed by AO/EB fluorescence staining.The results showed that streptomycin,uric acid and hydrogen peroxide all changed the morphological changes of cells,inhibited the proliferation of cells,increased the LDH release,and the results were concentration-dependent.In summary,streptomycin,uric acid,and hydrogen peroxide can induce rabbit RTECs to construct an adrenoceptive cell model in vitro.In the second part,we studied the protective effect of Huangkui Capsule-containing serum and Congrong Yishen Granule-containing serum on RTECstreated by three exogenous substances?streptomycin,uric acid and hydrogen peroxide?.The results showed that the high or medium dose of Huangkui Capsule-containing serum and the high or middle dose of Congrong Yishen Granule-containing serum can significantly reduce the inhibition effect of streptomycin cell proliferation and significantly decrease the LDH release of RTECs after streptomycin?P<0.05?.The high dose of Huangkui Capsule-containing serum and medium dose of Congrong Yishen Granule-containing serum can significantly reduce the inhibitory effect of uric acid on cell proliferation?P<0.05?.The high,medium or low dose of Huangkui Capsule-containing serum and the high or middle dose of Congrong Yishen Granule-containing serum can significantly decrease the LDH release of RTECs after uric acid?P<0.05?.The high dose,medium dose or low dose of Huangkui Capsule-containing serum and high dose of Congrong Yishen Granule-containing the general condition,blood biochemical indexes,urinary biochemical indexes,oxidative stress indexes and HE staining of renal cortex in normal group,model group and drug intervention group.The results showed that the weight of the model group was significantly lower than that of the normal group?P<0.05?,and the KI was significantly higher than that in the normal group?P<0.05?.The results showed thaturinary protein?UPT?,urinary creatinine?UCr?,serum creatinine?SCr?and blood urea nitrogen?BUN?were significantly higher in the model group than in the normal group?P<0.05?,and the clearance rate?CFR?of endogenous creatinine was lower in the model group than that in the normal group?P<0.05?.the activities of superoxide dismutase?SOD?and glutathione peroxidase?GSH-Px?in the renal cortex homogenate of the model group were lower than those in the normal group?P<0.05?.Compared with the model group,the KI of the drug intervention group was lower than that of the model group?P<0.05?.The serum creatinine?SCr?and urea nitrogen?BUN?in the Huangkui Capsule group were significantly lower than those in the model group?P<0.05?.The levels of urea nitrogen?BUN?and uric acid?UA?in Congrong Yishen Granule group were significantly lower than those in model group?P<0.05?.The activity of SOD and GSH-Px in Huangkui Capsule high dose group and Congrong Yishen Granule high dose group were significantly higher than those in model group?P<0.05?.In summary,the two kinds of anti-kidney prescription in vivo and streptomycin nephrotoxic formation of antagonism,reduce uric acid levels,improve renal tissue antioxidant capacity.In summary,Huangkui Capsule,Sunset Abelmoschus Flower,Congrong Yishen Granule and Cistanche deserticola Ma can protect the three exogenous substances?streptomycin,uric acid,H2O2?induced renal tubular epithelial cell injury in different degrees.Huankui Capsule and Congrong Yishen Granule have protective effect on streptomycin nephrotoxicity in rats,and can decrease uric acid and enhance the antioxidant capacity of renal tissue.Preliminary speculative mechanisms may be associated with antagonism of streptomycin and streptomycin-induced hyperuric acid and oxidative stress-induced renal tubular epithelial cell injury.
