Sunset Abelmoschus Flower is the dry corolla of Abelmoschus manihot(L.)Medic. Total flavones in the flowers of Sunset Abelmoschus is theeffective part,which can protect cerebral ischemia, myocardial ischemia andprotect the myocardial cells, improve blood rheology and anti lipidperoxidation. Huangkui dropping pill can treat cardiovascular andcerebrovascular disease is quick-acting solid preparation,which is prepared bythe traditional Chinese Medicine theory and according to modern preparationtechnology. We studied the extraction technology of total flavones in theflowers of Sunset Abelmoschus and purification technology of total flavonesby macroporous resin.We through orthogonal design to establish the optimumpreparation process of Huangkui dropping pill.Objects: To optimize the extraction technology of total flavonoids in theflowers of Sunset Abelmoschus;In order to improve the purity of totalflavones,we optimize purification technology of total flavones from theflowers of Sunset Abelmoschus by macroporous resin,and establish optimumtechnology parameters; In order to provide quick,convenient and stabledropping pills, orthogonal design is used to establish the optimum preparationprocess for Huangkui dropping pill which is prepared by solid dispersiontechnique.Methods:1The optimum acidification technology of the total flavonesfrom the flowers of Sunset Abelmoschus: The reflow time, the reflowtemperature, acidizing fluid volume and ratio of acidizing fluid on effect ofacidification of total flavonoids were evaluated by orthogonal design L49(3).2The optimum extraction technology of the total flavones from the flowers ofSunset Abelmoschus: The ethanol concentration, the extraction ethanolconsumption,extraction time and extraction times on effect extraction rate of total flavonoids were evaluated by orthogonal design L49(3).3The purificationtechnology of total flavones from the flowers of Sunset Abelmoschus bymacroporous resin: Spectrophotometric method was used to detect the totalflavonoids,the sample concentration,PH and ethanol concentration on effectthe rate of total flavonoids were evaluated by adsorptive ratio and desorptionratio.4The optimum preparation process for Huangkui dropping pill: Theformulation and molding technology of Abelmoschus manihot dripping pillwere optimized by orthogonal test with the proportion of matrix,types ofmatrix,dripping preparation temperature,the height of freeze-pillar as factorsand using variation coefficient,and appearance quality as index.Results:1The optimum acidification technology of the total flavonesfrom the flowers of Sunset Abelmoschus:25times the amount of acidizingfluid, the ratio of acidizing fluid was4:1,0.5h for reflow time, the temperatureof reflow was70℃.2The optimum extraction technology of the total flavonesfrom the flowers of Sunset Abelmoschus:12-folds of85%ethanol,2.5hours ofextraction,2times of extraction for extraction process.3The purificationtechnology of total flavones were obtained as follows: the sample is0.2g/ml,the sample quantity was3BV,the size ratio was1:6,the PH was5and75%ethanol was used as the spent regenerant.4The optimum preparationprocess for Huangkui dropping pill: ratio of drug to matrix was1∶3,matrixwas PEG4000and PEG6000(1∶0),temperature of dripping preparation was70℃,the height of freeze-pillar was120cm.Conclusions: The optimum extraction process for total flavonoids isstable and feasible;The macroporous resin can enrich the total flavoneseffectively;The Huangkui dropping pills made by this technology have goodproperties of shaping, prepared drop pills are uniform size and smooth roundshape,which is up to the quality requirements of drop pills.
