Neurodegenerative diseases have become one of the most common diseases with the development of the aging population.At present,the pathogenesis of neurodegenerative disease is basically confirmed.Most of the neurodegenerative diseases have the involvement of oxidative stress and inflammation in the course of the disease.Osthole as active ingredients extracted from Chinese herbal medicine,in recent years has been gradually research,existing research results show that osthole has effect on anti-inflammatory and anti oxidative stress,so we have reason to explore the effects of osthole on nerve cell function and mechanism.The experiment used two recognized typical neural cells--BV2 and PC12 cells.In order to ensure that the dosage of osthosin has no effect on the two kinds of nerve cells,we detected the activity of the cells by MTT.It was found that the osthole was not toxic to the cells at 4?g/mL,7?g/m L,and 10?g/mL.We used these three doses of osthole to pretreat cells 1h,then treated cells with LPS and H2O2 respectively to established cell inflammation and oxidative stress models.Subsequently,we detected the inflammatory and oxidative stress markers in the cells.It was found that osthole could reduce the expression of TNF-?,IL-6 and IL-1?,and played an anti-inflammatory role in this process.Meanwhile,we found that osthole could significantly reduce the expression of ROS,MDA and GSH,and adjust mitochondrial membrane potential,which also proved that osthole has the effect of anti oxidative stress.In order to further explore the mechanism of osthole to protect the nerve cells,we tested the treated cells by Western blot.It was found that osthole can increase the expression of Nrf2 and HO-1,and reduce the expression of P-I?B and P-P65.The results reveal that osthole activates the two signaling pathways of Nrf2 and NF kappa B to protect the cells.It can be seen that osthole can reduce the content of intracellular oxidation products,by regulating the Nrf2/HO-1 signaling pathway.At the same time,the production of inflammatory factors in the cell supernatant can be reduced by regulating the NF-kappa B signaling pathway.This study shows that osthole has the double effects of anti-inflammatory and antioxidant stress on the nerve cells.
