Glycosides From The Roots Of Securidaca Inappendiculata Hassk

Securidaca inappendiculata Hassk,a climbing shrub of the Polygalaceae family which belongs to the tribe Polygaleae,is mainly distributed in Yunnan,Guangxi,Guangdong,and Hainan provinces of southern China and the tropical regions of Asia.As a traditional Chinese herbal medicine,the roots of S.inappendiculataare were used as an anti-inflammatory,antibacterial,and antirheumatism agent.Compounds identified from S.inappendiculata previously include xanthones,oligosaccharide esters,saponins,benzophenones,hemiterpenoic acid glycoside,sterols and organic acids.Our previous antitumor constituents investigations of the aqueous portion of ethanol extract from the roots of S.inappendiculata have shown that the plant is a rich source for acylated triterpene saponins.As a part of continuing studies on the same portion,20 compounds were isolated by various chromatographic methods including silica gel,Sephadex LH-20,MPLC and preparative HPLC.On the basis of MS and NMR analysis 14 compounds were identified including seven lignan glycosides:acernikol-4??-O-?-D-glucopyranoside?SEC8?,?7R,8S?-dihydrodehydrodiconiferylalcohol9-O-?-D-glucopyranoside?SEC10?,?+?-syringaresinol-4-O-?-D-glucopyranosyl-?1?4?-O-?-D-glucopyranoside?SEC11?,?7R,8S?-5-methoxydihydrodehydrodiconiferylalcohol4-O-?-D-glucopyranoside?SEC 14?,?7R,8S?-dihydrodehydrodiconiferyl alcohol4-O-?-D-glucopyranoside?SEC 16?,?7R,8S?-dihydrodehydrodiconiferyl alcohol9?-O-?-D-glucopyranoside?SEC 17?,Glaberide I 4-O-?-D-glucopyranoside?SEC19?;three sucrose esters:3,6?-O-diferuloylsucrose?SEC 12?,arillanin A?SEC 13?,sibricose A5?SEC 18?;two hemiterpene glycosides:2-methene-butyric acid methyl ester-4-O-?-D-glucopyranoside?SEC2?,1-O-[6-O-feruloyl-1,5-anhydro-glucitol]-2-methene-butyricacidmethyl ester-4-O-?-D-glucopyranoside?SEC 7?;one benzyl benzoate:benzyl2-O-[?-L-arabinopyranosyl-?1"?6??-?-D-glucopyranosyl]-6-methoxybenzoate?SEC 6?;one organic acid ester:mehyl ferulate?SEC 1?.Compounds2,6,7 and 11are new compounds.Compounds 1,8,10,14,16-19 were isolated from the Securidaca genus for the first time.Cytotoxicities of compounds 2,6-8,10-13 and16 were evaluated against Lewis lung cancer A549,human cervical cancer Hela,and human breastcancer MCF-7 cell lines.Compounds 10 and 16 exhibited weak cytotoxic activities against Hela with IC₅₀0 values of 87.21,79.95?mol?L^-1.Compounds 10,13,and 16 showed weak cytotoxic activities against MCF7 with IC₅₀0 values of 89.75,88.41,and 59.98?mol?L^-1.
