| Objective Discuss effect of the function of spatial learning and memory and the activity of ChAT and AChE in the hippocampus of MID rats treated with "YiQiTiaoXue,FuBenPeiYuan" Moxibustion of Zhuang medicated thread.This study provides more scientific basis for the treatment of vascular dementia in traditional Chinese Medicine.We make the treatment of vascular dementia patients more convenient to offer advice and suggestions.Methods After modeling and screening,100 SD rats were randomly divided into the normal group,the sham operation group,the model group,the medicinal thread moxibustion group,the western medicine group and the non acupoint group,with six groups,10 rats in each group.The treatment of medicinal thread moxibustion group was "YiQiTiaoXue,FuBenPeiYuan" drug line point moxibustion,the western medicine group was gavage by donepezil solution,the treatment of non acupoint group was to use medicine line point moxibustion to stimulate the non acupoint point under two ribs,the remaining three groups were stimulated with catching with the same time of treatment(non real means of treatment).The course was four weeks.The spatial learning and memory ability of rats in each group before and after the intervention were detected by Morris water maze.ChAT and AChE activity in the hippocampus of rats was tested by colorimetric method.Results(1)Fixed position navigation test: Before the intervention,there were statistical differences between the model groups and the normal groups at the fourth day of training(P<0.05).The difference was further increased at the fifth day(P<0.01),and there was not statistical difference between the model groups(P>0.05).It shows that the model groups' abilities to acquire spatial information are significantly worse than that of the normal group.After the intervention,the medicinal thread moxibustion group and the western medicine group were significantly different from the model group at the fourth day of the training(P<0.05).The medicinal thread moxibustion group had a statistical difference compared with the model group at the fifth day(P<0.01),and there was also a statistical difference between the western medicine group and the model group(P<0.05),and there was not statistical difference between the medicinal thread moxibustion group and the western medicine group(P>0.05).It shows that the spatial information acquisition abilities of the medicinal thread moxibustion group and the western medicine group had been improved obviously.There was a statistical difference between the medicinal thread moxibustion group and the non acupoint group(P<0.01),and there was not statistical difference between the non acupoint group and the model group(P>0.05).It shows that the therapy of the medicinal thread moxibustion group has acupoint specificity.(2)Space exploration test: Before the intervention,the number of model groups passing through the original platform area was statistically different from that of the normal group(P<0.01).There were not statistical differences among the model groups(P>0.05).It shows that the abilities to preserve spatial information in the model groups are significantly worse than that of the normal group.After the intervention,the number of medicinal thread moxibustion groups crossing the original platform area was statistically different compared with the model group(P<0.05).It shows that the space information preservation ability of the medicinal thread moxibustion group has been obviously improved.There was a statistical difference between the medicinal thread moxibustion group and the non acupoint group(P<0.05),and there was not statistical difference between the non acupoint group and the model group(P>0.05).It shows that the therapy of the medicinal thread moxibustion group has acupoint specificity.There was not statistical difference between the western medicine group and the model group(P>0.05),and there was a statistical difference between the medicinal thread moxibustion group and the western medicine group(P<0.05).It shows that the medicinal thread moxibustion group is superior to the western medicine group in improving the space information preservation ability.(3)ChAT: The model group was significantly lower than the normal group(P<0.01).The medicinal thread moxibustion group was significantly higher than the model group(P<0.01).There was a statistical difference between the medicinal thread moxibustion group and the western medicine group(P<0.01).There was not statistical difference between the western medicine group and the model group(P>0.05).There was not statistical difference between the non-acupoint group and the model group(P>0.05).There was a statistical difference between the medicinal thread moxibustion group and the non acupoint group(P<0.01).It shows that the therapy of the medicinal thread moxibustion group can improve the activity of ChAT in the hippocampus of the model rats.The improvement effect is better than that of the western medicine group,and it has acupoint specificity.(4)AChE: The model group was significantly higher than the normal group(P<0.01).The medicinal thread moxibustion group was significantly lower than the model group(P<0.01).The western medicine group was significantly lower than the model group(P<0.01).There was not statistical difference between the western medicine group and the medicinal thread moxibustion group(P>0.05).There was not statistical difference between non-acupoint group and model group(P>0.05).There was a statistical difference between the medicinal thread moxibustion group and the non acupoint group(P<0.01).It shows that the therapy of the medicinal thread moxibustion group can improve the activity of AChE in the hippocampus of the model rats,and it has acupoint specificity.The improvement effect of the medicinal thread moxibustion group and the western medicine group is almost the same.Conclusion "YiQiTiaoXue,FuBenPeiYuan" Moxibustion of Zhuang medicated thread can improve the function of spatial learning and memory and the activity of ChAT and AChE in the hippocampus of MID rats,whose effect is better than donepezil.Moreover,this therapy has acupoint specificity. |
