Objective: In recent years,the incidence of prostate cancer significantly increase.It is of great significance to carry out the research on the molecular mechanism and interferential prevention of prostate cancer in China.At present,the treatment of prostate cancer mainly includes local treatment such as particle implantation and other localized prostate cancer radical treatment such as surgery,radiotherapy,chemotherapy,and take different comprehensive treatment according to the stage of cancer and the state of the patient.Due to the heterogeneity of prostate cancer,patients treatment options have been limited.This requires further study of the molecular characteristics of prostate cancer to identify the target of the drug,and seek natural plant medicine to reduce the toxic side effect of patient treatment.Lutein as a natural plant chemical has entered the people's view.It is widely found in plants containing more chlorophyll and is a carotenoid.Lutein has been found some effects such as antioxidant properties,enhance immune function,prevent and reduce the occurrence and development of cancer and other biological functions.This topic is based on human prostate cancer PC3 cells.We investigate the mechanism of lutein on the proliferation and apoptosis of PC3 cells in order to provide new theoretical guidance for the prevention and diagnosis of prostate cancer.Method:1.The effect of lutein on the activity of PC3 cells was detected by cck-8 method;2.The morphological changes of PC3 cells were observed after the treatment of lutein under an inverted microscope;3.The effect of lutein on the migration of PC3 cells was observed by cell scratches;4.The effect of lutein on invasion of PC3 cells was tested by transwell test;5.The effect of lutein on the cycle distribution of PC3 cells was detected by flow cytometry;6.The effect of lutein on apoptosis of PC3 cells was detected by flow cytometry(Annexin V-FITC/ PI double staining);7.The level changes of PC3 cells Bcl-2 m RNA,Bax m RNA treated with lutein were detected by RT-PCR;8.Western blot was used to detect the expression of apoptotic proteins Bcl-2,Bax in PC3 cells treated with lutein;Result:1.The results of CCK8 showed that lutein significantly inhibited the proliferation of PC3 cells after 24 h,48 h and 72 h,and showed a time and concentration dependence.2.It can be seen from the observation of cell morphology that the cell morphology significantly changes after 24 h with the increase of the concentration of lutein,and there is a tendency of obvious apoptosis.3.The experimental results showed that the drug concentration group could inhibit the migration of PC3 cells compared with the control group,and had a dose dependence with a significant difference in 160 ?g/ml(P < 0.01).4.Transwell experiment showed that the number of cells of lutein drug concentration group was less than that of the control group,and there was a dose dependence,and 160 ?g/ml was significantly different(P < 0.01).5.The results of flow cytometry showed that the cell growth cycle could be blocked in G0/G1 phase after the effect of different concentrations of lutein drugs in 48 h on PC3 cells.6.Flow cytometry results showed that the lutein drugs acting on PC3 cells after 48 h,the apoptosis rate increased significantly with drug concentration increased,when the drug concentrations of 120 ?g/ml,apoptosis rate up to 36.1%.7.The results of RT-PCR showed that lutein could lower the level of Bcl-2 m RNA at the transcription level and increase the level of Bax m RNA,and showed a dose dependence.8.Western blot experimental results showed that lutein could decrease the expression of Bcl-2 and promote the expression of apoptosis protein Bax,and showed a dose dependence.Conclusion:Lutein can inhibit the proliferation of PC3 cells in prostate cancer and promote apoptosis in prostate cancer.Its anti-tumor mechanism may be associated with blocking cell cycle,inhibiting cell migration,inhibiting cell invasion,regulating apoptosis-related protein expression and affect gene level.
