Background:Bronchial asthma is a chronic respiratory disease that occurs frequently in children in China in recent years.At present,the recognized traditional Chinese medicine for asthma in children is divided into three phases:attack stage,remission stage,and delaying stage,including eight types of syndromes:such as Phlegm heat resistance lung syndrome,Asthenia syndrome of lung and spleen and so on.The diagnosis of TCM syndromes in asthmatic children was diagnosed based on the experience of clinicians,there was no uniform standard,and the degree of international recognition was poor,in order to improve the standardization and objectivity of TCM syndromes,provide a basis for better dialectical treatment.In this study,metabolomic analysis of exhaled breath condensate in asthmatic children was conducted to investigate the syndrome nature of asthmatic syndrome in children.Objective:To study the differential metabolites of exhaled breath condensate in children with bronchial asthma,and to explore the metabolic syndromes of TCM syndromes in children with asthma.Methods:Fifteen children with phlegm heat resistance lung syndrome and 15 asthenia syndrome of lung and spleen during remission phase were collected.At the same time,15 normal children were collected as control group.Three sets of exhaled breath condensate samples were collected using the RTube exhaled breath condensate collection device.Multiple sets of samples were detected by GC-MS.After different metabolites were screened by statistical analysis,the changes in metabolic pathways were analyzed and the essential changes of asthmatic syndromes in different children were sought to be elaborated.Results:The results of metabolomics analysis of exhaled breath condensate showed that each syndrome group can be well distinguished.Differences in metabolites and related pathways have been found in the established models.Compared with the control group,the content of 1-phosphoglue,D-tagatose,5-aminopenta,2-ketobutyric acid and other substances in the exhaled breath condensate of children with phlegm-heat obstructing pulmonary syndrome was increased,and succinic acid was increased.The content decreased,the four metabolic pathways with higher correlation were starch and sucrose metabolism,lysine degradation,amino-sugar nucleotide sugar metabolism,and phenylalanine metabolic pathway.Lung-spleen-qi deficiency syndrome in remission period compared with the control group.We discovered the content of glucose 1-phosphate,pyroglutamic acid,L-isoleucine,threonine,and sarcosine in the exhaled breath condensate of children decreased.and the content of phenanthrene increased.Compared with the two syndromes,the content of mevalonolactone,L-Valine and phenanthrene was relatively low in the patients with Lung-spleen-qi deficiency syndrome.Conclusion:The GC-MS technique can better distinguish the difference between the sputum-heat-resisting lung syndrome and the lung-spleen-qi deficiency syndrome in the childhood asthma exacerbation period and the exhaled breath condensate metabolites in healthy children;between the two syndromes in different periods There are different metabolic differences and different metabolic pathway changes,and a preliminary interpretation of the metabolic patterns of asthma in children.
