Objective:To evaluate the effects of the water extract of Daphne genkwa about bradykinin and serotonin in scald rats by randomized controlled trials,and to investigate the analgesic effect and mechanism of the water extract of Daphne genkwa at the molecular level.Methods:Ninety SD rats were randomly divided into four groups:blank control group(group A),the water extract of Daphne genkwa group(group B)and HuangQin ointment group(group C),27 rats in each group.The remaining 9 rats served as normal control group.Deep second-degree scald pain model was made on the rat's back.The wounds were treated with physiological saline gauze,the water extract of Daphne genkwa gauze and Huang Qin ointment gauze.The pain threshold and level of each rat were determined at 3h,6h,and 12h after scald.as well as changes in bradykinin and serotonin levels in rat serum.RESULTS:1.The pain threshold and the pain level of rats in The water extract of Daphne genkwa group and Huang Qin ointment group were both decreased.Compared with the blank group,the difference was statistically significant(P<0.05);2.The water extract of Daphne genkwa group and Huang Qin ointment group had significant inhibition on the production of bradykinin and serotonin(P<0.05),and this inhibitory effect increased with the prolonged administration time;The inhibitory effect of bradykinin and serotonin of Huang Qin ointment group was more obvious than that in the water extract of Daphne genkwa group,and there was a statistically significant difference between groups(P<0.05).Conclusion:The water extract of Daphne genkwa can improve pain threshold,reduce pain level,and inhibit the production of bradykinin and serotonin in rat serum to some extent.Which can procide new ideas for our further analgesic mechanism research of The water extract of Daphne genkwa.
