Objective:The aim of this study was to find out the correlation between the fingerprint of valotile oil from Curcuma kwangsiensis S.G.Lee et C.F.Liang and its efficacy.Moreover the paper may provide the new ways to clarify the basis of its effective substance,set up its quality control and therapeutic efficacy assessment standards.Methods:1.The L9?34?orthogonal test and the yield of volatile oil was used to optimize the extraction process of Curcuma kwangsiensis S.G.Lee et C.F.Liang valotile oil by steam distillation.2.The GC-MS analysis and“Computer Aided Similarity Evaluation System”?recommended by the National Pharmacopoeia Committee?which is2012 version were used to compare the similarities and differences of valotile oil from 10 batches of Curcuma kwangsiensis S.G.Lee et C.F.Liang and compute the similarity,which were established the fingerprint and acquired the characteristic peak area.3.The tumor cells were treated with valotile oil at different concentration?10,20,50,100?g/ml?,negative control and blank group.The effect of inhibiting proliferation were detected by CCK-8 method,including selecting of sensitive tumor cell lines from HepG2 hepatoma cells,NCI-H460lung cancer cells and PC12 Rat Pheochromocytoma.4.The characteristic peak area and inhibiting proliferation rate to HepG2hepatoma cells were analyzed by grey relational methods,and the original data was standardized.Using the standardized data,the correlation coefficient and correlation of the characteristic peak and inhibiting proliferation rate to tumor cells and the absolute difference of reference series were all calculated.Results:1.The best extraction process of steam distillation of water of volatile oil was added to the sample 100g,with 6 times the amout of water,heating extraction 7h,adding sodium chloride 50g.2.This volatile oil consists of 83 components by the GC-MS detection,of which 51 compounds were identified,mainly sesquiterpenes and their derivatives,identified 30 peaks from 10 batches of Curcuma kwangsiensis volatile oil,the peak area and retention time were in line with the requirements of traditional Chinese medicine fingerprint technology required,and the similarity of 10 batches were between 0.91.0,found no outliers,meet the requirements of chromatographic fingerprint of traditional Chinese medicine.3.After dose of 48h,different concentrations of essential oil on the growth of HepG2 hepatocellular carcinoma cells were significantly inhibite proliferation.The comparison of concentrations showed that the proliferation inhibition of 20,50 and 100?g/ml concentration groups increased significantly compared with those of 10?g/ml concentration group.The results showed that Curcuma kwangsiensis essential oil can inhibit the proliferation of HepG2 cells both in a dose effect dependent manner,and in a time dependent effect relationship.4.The first of the association was curcumenol,which has a high correlation with the inhibition of hepatoma cells,with a correlation of 0.88±0.06.Andthenextstepwascurcumenol,curcumenone,?-cyclodihydrocostunolide,germacrone,?-elemene,which correlation degree were above 0.70.In addition,the correlation of other 20 characteristic peaks was between 0.60-0.70.Conclusion:1.Adding sodium chloride co uld improve the extraction rate of volatile oil of Curcuma kwangsiensis.2.The components which were in GC-MS fingerprint of 10 batches of Curcuma kwangsiensis volatile oil were separated,so this method could be used as a quality control method of Radix Curcuma kwangsiensis.3.Curcuma kwangsiensis volatile oil had obvious inhibitory effect on HepG2 cells.The next step is to further clarify its mechanism of action.4.The antitumor effect of turmeric essential oil on liver cancer cell line HepG2 was its"effective group"common result,this study determined the Curcuma kwangsiensis volatile oil"group"fingerprint effect on the size of the contribution,which provided new ideas and methods for the establishment of Chinese medicine"spectrum effect Association"fingerprint,clarifying pharmacodynamic material basis,developing quality control standard of Chinese traditional medicine and efficacy evaluation.
