Introduction: The incidence of infertility in the society is increasing,and the reasons are complicated.Because of the aggravation of environmental pollution,bisphenol A(BPA)as a reproductive toxic substance has attracted the attention of scholars.Objective: To explore the association of expression level of cystic fibrosis transmembrane conductance regulator(CFTR)and the activity of mouse sperm exposed to high level of bisphenol A(BPA),Therefore,it provides theoretical support and guidance for the treatment of male infertility in clinical work.Methods:Mouse sperm were incubated with BPA at doses of 0?g/mL,10?g/mL,100?g/mL,500?g/mL.The sperm survival was evaluated by the eosin-nigrosin staining,capacitation and acrosome reaction was measured with chlortetracycline staining,and the protein expression levels of cAMP,phosphorylated CREB(p-CREB)and CFRT were analyzed with Western blotting.Results: BPA exposure at the varied doses significantly decreased the survival rate,capacitation rate and acrosome reaction rate,even over 50% decreases on the activities were observed at the 500?g/mL group.The quantitative expression levels of cAMP,p-CREB and CFTR in mouse sperm were also dramatically reduced,and over 70% decreases were observed at the 500?g/mL group.CFTR levels were strongly correlated with expression levels of cAMP and p-CREB,and rate of capacitation.Conclusions: BPA decreased the activity of mouse sperm,and CFTR was possibly related to the adverse effects of BPA on sperm.
