Objective:This study discusses the etiology and pathogenesis of AD from Tujia Medical Theory,and the theoretical basis of Bunao Gandu Fang in the treatment of AD.Finding the possible mechanism of this prescription for the treatment of AD.In order to provide extensive experimental evidence for Tujia medicine on clinical,and discover a new method for curing AD by using traditional medical treatment.Methods:After feeding 2 weeks,divide 70 healthy male SD rats randomly into 7 groups,make sure each group has 10 rats,and 7 groups named:the normal group,the model group,the compound group,the Baisanqi group,the Water calamus group,the yanzelan group,and the Anshen group.Except for the normal group,all six groups were injected into the hippocampus to produce the AD model.After the model wounds were restoring basically,the rats in each group were treated with gastric lavage.The five drug groups were given the corresponding drug gavage,and the normal group and the model group were given the same volume of physiological saline for 28 days.Observing the effect of Bunao Gandu Fang on the behavior of AD rats,the effect of Bunao Gandu Fang and its decomposed prescriptions on the morphology of hippocampus in AD rats subjected to HE staining and Nissl staining.What's more,observing the effect of Bunao Gandu Fang and its removal on the contents of IL-1??IL-6?TNF-a in serum of AD rats by using ELISA method,the immunohistochemical method as well as PCR method.Besides,through WB method,observing the effects of the Bunao Gandu Fang and its removal on the PI3K/AKT,MAPK signaling pathway in the hippocampal tissue of AD rats and the effects of il-1,il-6,and tnf-alpha in the hippocampal tissue of AD rats.Results:The behavioral observation of rats showed that the study and memory function of AD rats was significantly improved compared with the model group after the treatment of pneumoconiosis.Through ELISA method,and the results showed that the Bunao Gandu Fang was effective in reducing the content of inflammatory factors.Through PCR method,the immunohistochemistry method,and WB method,Bunao Gandu Fang has been found the function to reduce the level of IL-1??IL-6?TNF-ain the brain tissues of AD rats.The experimental results of the signaling pathway showed that the p3k/AKT and P38 MAPK signaling pathway may be activated by the PWP.It is statistically significant that the experimental results of serology,protein detection and signaling pathway showed that the drug effect of compound prescription was better than that of the disassembly.Conclusions:Both Bunao Gandu Fang and its decomposed prescriptions can effectively improve the learning and memory function of AD rats,and show obvious curative effect on AD-related biochemical indicators.AD can be obtained through Tujia medicine for brain encephalopathy Good remission;Exploration of signal pathways The experimental results show that Bunao Gandu Fang may attenuate neuroinflammation and protect brain function by activating PI3K/AKT and P38 MAPK signaling pathways and inhibiting ERK MAPK and JNK MAPK signaling pathways.
