Objective:To investigate the role of IL-6-IL-17-CD13 signal regulation axis in renal damage of essential hypertension.Methods:1.Weight and blood pressure were measured in SHR rats and their wild type control WKY rats at 4,6,10,and 30 weeks,respectively;and concentration of IL-6,IL-17 and TGF-?1 in plasma was detected by ELISA;relative expression of IL-6,IL-17,CD13,CD4,RORyt,TGF-?1 mRNA in the kidney was detected by RT-PCR;expression of IL-6,IL-17,CD13,CD4,RORyt,TGF-?i,a-SMA and Collagen-I in the kidney was detected by immunohistochemical staining;level of renal fibrosis-was detected by Masson staining.2.SHR rats were randomly divided into IL-6 monoclonal antibody blocking group(SHR-B group)and normal saline control group(SHR-C group).The rats in the SHR-B group were intraperitoneally injected with IL-6 monoclonal antibody 10 ?g/day,the rats in the control group were intraperitoneally injected with equal volume of normal saline,for a total of 14 days.The above indicators were dynamically detected at 4,6 and 30 weeks,respectively.Results:1.The weight of SHR rats was lower than that of WKY rats at the same time,there was a significant difference between the two groups at 30 weeks(P<0.05);the systolic pressure of SHR rats was significantly higher than that of WKY rats at 6,10,30 weeks(all P<0.05);the concentration of IL-6,IL-17 and TGF-?1 in plasma of SHR rats was also significantly higher than that of WKY rats in the same time,with a difference between the two groups(all P<0.05);the relative expression level of IL-6,IL-17,CD13,CD4,RORyt and TGF-?11 mRNA in the kidney of SHR rats was also significantly higher than that of WKY rats in the same time at 6,10,30 weeks(P<0.05 or P<0.01);the immunohistochemistry average optical density of IL-6,IL-17,CD13,TGF-?1 a-SMA and Collagen-I in kidney of SHR rats was also significantly higher than that of WKY rats in the same time at 6,10,30 weeks(P<0.05 or P<0.01);Masson staining showed that compared with WKY rats,the degree of renal fibrosis in SHR rats increased significantly,and there was significant difference between the two groups at 10,30 weeks(P<0.05).2.Compared with SHR-C rats,the systolic pressure of SHR-B rats was decreased significantly at 30 weeks(P<0.05);the concentration of IL-17 and TGF-?1 in plasma of SHR-B rats was also significantly decreased at 6,30 weeks,compared with SHR-C rats(P<0.05);the relative expression of IL-17,CD13,CD4,ROR?t and TGF-?1 mRNA in the kidney of SHR-B rats was also significantly lower than that of SHR-C rats at 30 weeks(P<0.05);the immunohistochemistry average optical density of IL-17,CD13,TGF-?1 a-SMA and Collagen-I in kidney of SHR-B rats were also significantly lower than that of SHR-C rats in the same time at 30 weeks(P<0.05 or P<0.01);Masson staining showed that compared with SHR-C rats,the degree of renal fibrosis in SHR-B rats was decreased,and there was significant difference between the two groups at 30 weeks(P<0.05).Conclusion:The IL-6-IL-17-CD13 signal regulation axis plays an important role in renal damage of essential hypertension;at the beginning of the formation of hypertension,blocking the IL-6-IL-17-CD13 signal regulation axis can reduce or delay the procession of hypertension and renal damage.
