Objective:The effect of Guanxin Xiaoquan capsule on the expression of IL-8,IL-10 and CD105 in atherosclerosis rabbits,To explore the role and possible mechanism of Guanxin Xiaoquan capsule in inhibiting the development of atherosclerosis,Provide theoretical basis for clinical prevention and treatment of coronary heart disease.Methods:40 Japanese white rabbits were divided into blank control group,high fat model group,Guanxin Xiaoban Capsule group and simvastatin group according to the random number table grouping method.10 in each group.Except for the blank control group,the other groups were given high fat diet 100 g every morning,Establishment of atherosclerotic model for 12 weeks.After the success of the modeling,the other 3 groups,except the blank control group,continued to give high fat feed 100 g daily,Guanxin Xiaoquan capsule group gavage of 160 mg per kilogram daily,Simvastatin group gavage 0.58 mg per kilogram daily,The blank group and high fat model group were gavage 5m L normal saline daily,After 4 weeks of continuous feeding,Pathomorphological observation of aorta of atherosclerotic rabbits,The expression of IL-10 in atherosclerotic plaques by immunohistochemical staining,Detection of CD105 expression by immunofluorescence,Detection of IL-8 and IL-10 levels in serum by ELISA,Enzyme method applied for examing blood lipid.Results: 1 Detection of blood lipid by Enzyme method: Compared with model group,the content of LDL-C in the serum of Guanxin Xiaoquan capsule group was significantly reduced(P<0.01),a significant increase in serum HDL-C content(P< 0.01).2 inspection result of pathomorphology: In the model group,the intima of the aorta was thickened,the intima surface of the aorta was less smooth,the endothelial cells were damaged,the inner membrane thickened,and the atherosclerotic plaques were more obvious.In the control group,the thickness of the aortic wall was more uniform,and the intima was smooth and complete.The part of the endometrium was thicke ned and the endothelial cells were swollen and denatured in Guanxin Xiaoban Capsule group and simvastatin group,compared with the model group,the foam cell and the lipid deposition decreased in varying degrees.3 Immunofluorescence results of CD105 tissue: No positive expression of CD105 was found in the blank control group.A large number of CD105 expressions in the model group.A small amount of CD105 positive expression was found in the simvastatin group,A slight thickening of the intima,But lighter than m odel group.A small amount of CD105 expression in Guanxin Xiaoquan capsule group,Smooth and no obvious thickening of the intima of the arteries.4 Determination of the content of IL-8 and IL-10 in serum by ELISA: Compared with the control group,the content of IL-8 in the serum of the model group was significantly increased(P< 0.01).The content of IL-8 in Guanxin Xiaoban Capsule group and simvastatin group was less than that of the model group,and the difference was statistically significant(all P<0.01).Compared with the control group,the content of IL-10 in the model group was significantly lower than that of the control group(P<0.01).The content of IL-10 in the serum of Guanxin Xiaoban Capsule group and simvastatin group was higher than that of the m odel group,the difference was significant(all P< 0.01),The content of IL-10 in Guanxin Xiaoquan capsule group was significantly higher than that of simvastatin group.5 The average optical of IL-10 concentration in tissue : Compared with the control group,the expression of IL-10 in the aortic wall of the model group was significantly reduced(P<0.01);Compared with the model group,the expression of IL-10 in the Guanxin Xiaoban Capsule group and the simvastatin group increased significantly(all P< 0.01),But Guanxin Xiaoban Capsule is superior to simvastatin in increasing the content of IL-10(P<0.01).Conclusions : 1 Guanxin Xiaoban Capsule can postpone the progression of atherosclerotic plaque by regulating lipid metabolism and protect vascular endothelial cells.Guanxin Xiaoban Capsule could Anti atherosclerotic plaque by reducing the expression of inflammatory factor IL-8,improving the anti inflammatory factor IL-10 and Strengthening the anti inflammatory response of atheroscl erotic plaque.2 One of the mechanisms of Guanxin Xiaoban Capsule to stabilize atherosclerotic plaque may be through inhibition of the expression of CD105 in atherosclerotic plaques and reduction of pathological angiogenesis in atherosclerotic plaques.
