Objective:To research the extraction technology of γ-lin-olenic-the main constituent of borage officinalis L.,then to un-derstand and master the making method of borage effervescent tablets.To establish a method for the determination of the content of γ-linolenic-the main constituent of borage and in order to provide scientific basis for the further development and utilization of borage we will observe the antifatigue effectof borage and explore the mechanism of its function.Methods:(1)To prepare the water extract of the borage and extract the whole plant components.By comparing the total extraction rate and the extraction content,the best extraction technology of the water extract of borage was selected.For the last,the flake shape,weight difference,disintegration time and foaming amount of the effervescent pieces were analyzed.(2)Using the method of gas chromatography-mass spectrogr-aphy,we determine the main ingredient of borage γ-linolenic a-cid content preliminaril.(3)To establish the fatigue model of mice,and then the act-ivities and contents of lactate dehydrogenase(LDH)-lactic acid(LA),blood urea n itrogen(BUN)in plasma and hepatic glycogen in liver of fatigue mice were determined,at the last the mech-anism of antifatigue effect of borage was analyzed.Results:(1)The optimum extraction conditions of borage extract was to soak one hour and then to extract for one hour,calculate the extract rate was 2.35%,and we should make sure that the flake shape,weight difference,disintegration time and foaming amount of the effervescent tablets were all in accordance of the regulations of Pharmacopoeia.(2)The time of linolenic acid methyl ester’s peaking was 13.60min,molecular ion peak were 32 and 79.The determination results for γ-linolenic acid’s content in borage plant was 0.65mg/g,the content in the powder of borage water extract was 0.40ing/g,and the content of borage effervescent tablets is 0.098mg per tablet.(3)The high dose group of borage effervescent tablets could significantly increase the activity of LDH in plasma of mice(P<0.05),and decreased the content of LA in mice significantly(P<0.05),and significantly decreased the plasma BUN content in mice(P<0.05).In the medium and low dose groups,the activity of LDH and the content of liver glycogen were increased and the contents of LA and BUN were decreased,but the effect was not obvious.Conclusion:(1)The best extracting tne of water extract and the optimum ratio of effervescent tablets were determined.(2)We usded the methods of gas chromatography-mass spect-rometry(GC/MS)to make sure that the borage γ-linolenic acid is contented in 0.65mg/g.(3)It was concluded that the borage effervescent tab let had a certain anti-fatigue effect,which might be related to the increase of lactate dehydrogenase activity,decomp-osition and removal of lactic acid and urea nitrogen after exercise.
