Objective To investigate the effect and mechanism of berberine hydrochloride in the adjuvant treatment of ulcerative colitis(UC).Methods The rats were divided into 5 groups(n=8),of which four groups were given concentration of 5.5% dextran sulfate sodium(DSS)solution to drink freely.The remaining group was given distilled water to drink freely for seven days,forming a UC rat model.The next seven days,rats were stopped drinking DSS solution,and all experimental groups rats were given distilled water to drink freely.At the same time,normal control group and DSS model group were given by gavage with distilled water,mesalazine group was given by gavage with mesalazine(420mg/kg),berberine hydrochloride group was given by gavage with berberine hydrochloride(100mg/kg),and berberine hydrochloride combined with mesalazine group was given by gavage with berberine hydrochloride(100mg/kg)combined with mesalazine(420mg/kg).The degree of disease activity(DAI score)in five groups of rats and the pathological changes of colon tissue in each group were observed.Levels of serum IL-9 were measured by ELISA,and the reverse transcription polymerase chain reaction(RT-PCR)and immunohistochemistry were used to detect the expression levels of TLR2 mRNA and TLR2 protein in rat colon tissues.Results 1.Compared with the normal control group,the DAI score,the colonic histopathology score,the IL-9,the TLR2 mRNA and TLR2 protein expression in the DSS model group were significantly higher(P < 0.01).2.Compared with the DSS model group,the fourteenth day DAI score,serum IL-9 expression of mesalazine group and berberine hydrochloride group decreased(P < 0.05).Compared with the DSS model group,the colonic histopathology score of mesalazine group decreased significantly(P < 0.01).Compared with the DSS model group,the fourteenth days DAI score,colonic histopathology score and serum IL-9 expression of berberine hydrochloride combined with mesalazine group were significantly decreased(P < 0.01).Compared with the mesalazine group and berberine hydrochloride group,the colonic histopathology score of berberine hydrochloride combined with mesalazine group decreased(P < 0.05).3.Compared with the DSS model group,the expression of TLR2 mRNA and TLR2 protein in the colon tissue of mesalazine group,berberine hydrochloride group,berberine hydrochloride combined with mesalazine group was significantly decreased(P < 0.01).Compared with the mesalazine group and berberine hydrochloride group,the expression of TLR2 mRNA and TLR2 protein in the colonic tissue of berberine hydrochloride combined with mesalazine group decreased(P < 0.05).Conclusion Berberine hydrochloride may enhance the effect of methalazine in the treatment of ulcerative colitis by reducing the level of IL-9 and the expression of TLR2 mRNA and TLR2 protein.
