Objectives : On the basis of analysing the characteristics of glucose and lipid metabolism and the level of sex hormone in patients with polycystic ovary syndrome(PCOS),animal models of PCOS were established by modified Poretsky method and letrozole gavage method to determine which one is closer to the metabolic and endocrine characteristics of PCOS patients.Then this study analyzes the expression of PPAR ?,?,? in ovarian tissues of peroxisome proliferator activated receptor(PPAR),and to explore the relationship between this three subtypes and abnormal metabolism of PCOS.Methods:Using the PCOS diagnostic criteria issued by the Ministry of Health in2011,thrity-three patients with PCOS were collected from gynecological and endocrine clinics in Zhongshan Hospital affiliated to Dalian University between May 2016 and February 2018.Height,weight,waist circumference,hip circumference and waist-to-hip ratio were recorded.Serum glucose metabolism indexes and lipid metabolism index were measured by automatic electrochemiluminescence assay.Thirty healthy women were taken as control group,and the above indexes were measured and recorded at the same time.The characteristics of glucose and lipid metabolism and sex hormone levels in patients with polycystic ovary syndrome(PCOS)were analyzed by software SPSS21.0.Modeling of PCOS by modified Poretsky method(group A): SD rats were given subcutaneous injection of Novoline N into the neck of rats with 2.0 IUU once a day for21 days.Three IUU HCG(dissolved in 0.2ml saline)was injected in to rats twice a day.Modeling of PCOS by Letrozole group(group B): Rats were given letrozole 1 mg / kg once a day for 21 days after being dissolved in 1% hydroxy methyl cellulose solution once a day.Rats in group C of blank control group were given normal food and waterdaily.The changes of estrus cycle were observed by vaginal smears,and the body weight and ovarian weight of rats were measured.The changes of ovulation and follicle in rats were observed by ovarian tissue section.Immunohistochemical method was used to detect the expression of three subtypes of PPARs in the ovarian tissues of experimental animals.Results:In this study,thrity-three patients with normal weight in the PCOS group accounted for 69.7%(23/33);overweight or obesity accounted for 30.3%(10/33),there was no low weight case;90.1%(3/33)patients existed insulin resistance,27.27%(9/33),and the incidence of abnormal blood lipid was 15.15%(5/33),and the incidence of MS was 3.03%(1/33).T,LH,LH/FSH ratio in the PCOS group raised and the difference was statistically significant(P < 0.05)compared with the control group(P < 0.05).The FPG,FINS,HOMA-IR in the PCOS patient group was significantly higher than the control group,and the difference was statistically significant(P < 0.05).The results of animal experiments showed:(1)compared with the control group,the weight of rats and ovary increased in the letrozole group and the improved Poretsky group,and the change in the modified Poretsky group was more obvious.(2)the level of T,FINS,HOMA-IR in the group of letrozole was higher than that of the control group(P < 0.05),but LH,FSH,E2,P value,LH/FSH ratio was no significant difference(P > 0.05).The T,LH level,LH/FSH ratio,FPG,FINS,HOMA-IR,TG of the improved Poretsky group showed significantly higher than the control group and the FSH reduced statistically significantly compared with the control group.(3)HE staining showed that both improved Poretsky group and letrozole group had polycystic changes of ovary,the follicle cavity was cystic enlargement and the number of granular cell layers decreased.(4)the immunohistochemical detection results showed that the three subtypes of PPAR ?,PPAR ? and PPAR ? were expressed in the ovarian tissue.Compared with the blank group,the ovarian tissue of the modified Poretsky group was PPAR ?,?.Conclusion:1.The incidence of obesity,impaired glucose tolerance,diabetes mellitus,dyslipidemia,metabolic syndrome were higher than that in the normal control group,the sex hormone TH?T?LH / FSH levels were higher,and FSH become lower in PCOS patients.The changes were similar to those reported in the literature and related guidelines.2.The metabolic and endocrine features of the modified Poretsky method were closer to those of PCOS patients than the letrozole method.It can be used as a better modeling method for subsequent experiments.3.Two of the three subtypes of PPARs showed high expression in the ovarian tissues of the rat model induced by modified Poretsky method,which may be related to the abnormal metabolism of PCOS.
