Objective:To observe mRNA and protein expression of MAP-2, NF-L in rat hippocampus of cerebral ischemia-reperfusion model by XNKQ acupuncture on; explore the protection mechanism by XNKQ acupuncture on cerebral ischemia-reperfusion injury.Methods:SD rats were divided into normal group, sham-operation group, model group, XNKQ acupuncture group (acupuncture) and Meridian traditional acupuncture group (control group), with improved suture method Longa focal cerebral ischemia-reperfusion model,to observe rats’neurological score changes after acupuncture intervention, while the use of Real-time PCR and Western blot were detected in the hippocampus of rats MAP-2, NF mRNA and protein expression.Results:(1) neurological score:to compare with he model group,the acupuncture group and the control group,the difference neurological score was statistically significant (P<0.01) after treatment; the difference neurological score before and after treatment of acupuncture group and control group was significant statistical significance (P<0.01); the difference neurological score of acupuncture group and control group was no significant difference after treatment (P>0.05).(2) mRNA expression of MAP-2and NF-L:The acupuncture group and the control group compared with model group MAP-2, mRNA expression of NF-L significant difference was statistically significant (P<0.01); acupuncture group and the control group MAP-2, mRNA expression of NF-L difference was statistically significant (P<0.05).(3) protein MAP-2and NF-L:The acupuncture group compared with model group MAP-2, NF-L protein expression was significant difference was statistically significant (P<0.01); the control group and model group Compare MAP-2, NF-L protein expression differences were statistically significant (P <0.05); acupuncture group compared with control group MAP-2, the protein expression of NF-L difference was statistically significant (P<0.05).Conclusion:XNKQ acupuncture and traditional acupuncture may improve neural function after cerebral ischemia-reperfusion injury in rat, but the experiment has not been able to show that whichever is more dominant. XNKQ acupuncture can improve more significantly the hippocampus of rats MAP-2, NF mRNA and protein expression than traditional acupuncture, so as to better reduce ischemic brain acupuncture tissue damage by stable cytoskeleton. Regulate cytoskeletal stability may be play an important role in brain protection for XNKQ acupuncture treatment.
