As a kind of traditional tea,Coreopsis tinctoria Nutt.has been used for cardiovascular disease prevention,such as hypertension,hyperlipidemia and so on.In this study,the flavonoids of Coreopsis tinctoria Nutt.were studied systematically.And the quality evaluation criteria of Coreopsis tinctoria Nutt.from different sources was established.1.Study on the flavonoids of Coreopsis tinctoria Nutt.16 compounds were isolated and identified from methanol extract of Coreopsis tinctoria Nutt.by a variety of separation methods combining with moderm chromat-ographic and spectrum technique.These compounds were quercetin,quercetin-7-O-?-D-lucopyranoside,5,7,4'-trihydroxyflavone,2R/S-5,7,3 ',5-tetrahydroxyflavanone-7-O-?-D-lucopyranoside,isookanin,luteolin,marein,butein,flavanomarein,taxifolin,butein-4'-O-?-D-lucopyranoside,5,7,3',5'-tetrahydroxyflavanone,okanin,sulphuretin,6,7,3',4'-tetrahydroxyaurone,and linocinnamaxin respectively.2.Optimization of the extraction conditions of flavonoids of Coreopsis tinctoria Nutt.Orthogonal design method was used for investigating the effects of temperature,size and extraction solvent on the total yield of marein and flavanomarein with immersion extraction method and heating reflux method respectively.The total content of marein and flavanomarein was determined by HPLC to optimize the sample pretreatment conditions of Coreopsis tinctoria.The best pretreatment method was that samples were smashed,then heated reflux in methanol at 70? for an hour,and repeated 3 times.3.Quality evaluation of Coreopsis tinctoria Nutt from different sourcesTen compounds were selected as the representatives for the flavonoids in Coreopsis tinctoria.Then their contents were analysed by HPLC respectively.And some conclusions were obtained.(1)The contents of flavonoids in Coreopsis tinctoria from different sources were quite different.So it was very necessary to evaluate their quality and eatablish their corresponding grade standard.(2)According to the results of the content determination,the quality of samples XJ-8(wild source in Hetian Pishan Kerrey Yang area)and XJ-7(cultivated source in Dabancheng area)were best of all.Thei totalr content of flavonoids was 86.02 ± 0.12 mg/g and 84.90 ± 0.16 mg/g respectively.(3)Quality evaluation criteria was established.The total content of marein,flavanomarein and isookanin was taken as evaluation indicator.The sample whose content more than 70.00 mg/g was superior.The sample whose content between 52.00 mg/g and 70.00 mg/g was middle.The sample whose content less than 52.00 mg/g was the lowest.
