Coreopsis tinctoria Nutt.,which is a traditional tea,is widely cultivated in Xinjiang.It is commonly known as Xueju.Flavonoids are a major type of natural active substance,and they are low toxic.Because of the multiple effects of flavonoids,this study explored the anti-aging effect of five main flavonoids in Coreopsis tinctoria Nutt.,which used Caenorhabditis elegans as experimental object and the five flavonoids are marein,flavanomarein,isookanin,6,7,3',4'-tetrahydroxyaurone and quercetin.The results are as follows:1.Flavonoids in Coreopsis tinctoria Nutt.can prolong the lifespan of wild type C.elegans N2.By comparing the lifespan of N2 in different groups which feeded with four compounds other than quercetin,we can determine whether these compounds can delay aging process of N2.The results showed that compared with the vacant control,the survival curves of N2 feeded with these compounds shifted to the right,and the mean and maximum lifespan were higher than those of the blank control,which with statistically significant differences.2.Flavonoids in Coreopsis tinctoria Nutt.can enhance the anti-oxidant ability of N2.Dealing N2 with H2O2,and the survival rate of N2 in different treatment groups were compared,by which can determine whether these compounds had antioxidant effect in vivo.The results showed that when N2 were on acute oxidative stress,the survival rate of N2 treated with different compounds were higher than that of the control group,and there was a significant difference,which suggested that these compounds had antioxidant effects.By comparing the relative proportion of DAF-16 nucleus translocation cells,this study explored whether the anti-oxidant activity of the five active compounds was related to the promotion of DAF-16 transfering into the nucleus.The results showed that,compared with the vacant control,DAF-16 nucleus translocation rate in C.elegans treated with the compound increased,suggesting that the antioxidant effect of these compounds was related to the promotion of DAF-16 transfering into nucleus under oxidative stress.3.Flavonoids in Coreopsis tinctoria Nutt.can reduce intestine fat accumulation in N2.Intestine fat accumulation model of C elegans was established by feeding N2 whith glucose.Low concerntration of Nile Red in vivo staining was used to stain the intestine fat and determine whether the five compounds had lipid-lowering effect.The results showed that compared with the blank control,the fluorescence intensity in C elegans which fed with glucose was stronger,indicating that the model was successfully established.Compared with the model group,fluorescence intensity in C.elegans intestine which treated with five compounds was decreased,indicating that these compounds had lipid-lowering effect.By analyzing the expression of lipid metabolism related genes and protein in C.elegans,we found 50 ?M marein can significantly reduce the expression of FAT-7.The genes expression did not show any changes,which were lipid synthesis genes fat-7,sbp-1 and lipid degradation gene nhr-49,but the expression of FAT-7 was decresed,which suggested that marein can decrease lipid in C.elegans intestine by reducing the expression of FAT-7.In summary,the five flavonoids in Coreopsis tinctoria Nutt.can prolong the lifespan of C.elegans,and increase the anti-oxidant ability of C.elegans under acute oxidative stress by promoting DAF-16 nucleus translocation.In addition,marein can reduce intestine lipid in C.elegans by reducing the expression of FAT-7.Therefore we can conclude that the five flavonoids in Coreopsis tinctoria Nutt.have good anti-aging activity.
