Objective:To explore the resistance to macrolide drugs in the local syphilis by detecting the mutant loci of the resistant gene of Treponema pallidum Tp to the macrolide drug in Xinjiang,and to provide the basis for the treatment of syphilis with macrolide drugs.Methods:204 cases of first and two stage syphilis patients were collected from March 2016 to October 2017 at the First Affiliated Hospital of Xinjiang Medical University.The serological indexes met RPR positive,and the titer was more than 1:8 and TPHA was positive.The TP genome DNA was extracted from all the collected specimens and the TP specific gene PolA was amplified by PCR amplification.The positive specimens were amplified by PCR nested 23SrRNA resistant gene,and the amplified product MboII,BsaI restriction endonuclease was used to confirm whether there was a resistance mutation site.Results:27 specimens of 204 samples collected in this experiment were amplified by TP specific gene PolA,the amplification rate was 13.24%,of which 13 were Han,7 in Uygur,6 in Kazak,1 in other minority groups,and 27 positive samples were 23SrRNA positive,and the detection rate of resistance genes was 100%.There are A2058G resistant mutations in 24 cases and A2059G resistance mutations in 3 cases.Conclusion:The TP strains in Xinjiang area are generally resistant to macrolides.Macrolides are not recommended for the treatment of syphilis.
