Thuja sutchuenensis is known as Yabai Tree or Sichuan Cebai.It is an extremely endangered species in the world,and it is a unique species of China.It belongs to Cupressaceae Thuja.Currently remaining wild plants are very few,and they are only dicovered on the steep slope and cliff far away from residential areas in southwest China.It could survive thousands of years with strong self-healing ability.Not only is it very resistant to decay,but also it has strong antioxidant activity.And it is an ancient Chinese herbal medicine.Used some chromatographic column methods of Silica gel column,RP-18 reversed-phase silica gel column,Sephadex LH-20 gel column,Macroporous resin adsorption chromatography,HPLC Methods etc.in this research,which to extract chemical separation of the cultivation T.sutchuenensis branch of Qingyuan in Guangdong province.And other techniques of NMR,Mass Spectrometry etc.were also used to identificate chemical structure.Then 12 kinds of compounds have been separated and identified:(1*)thuja phenol A((E)-8-methoxy-1,1,4atrimethyl-6-styryl-2,3,4,4a,9,9a-hexahydro-1H-xanthene),(2)3,5-dihydroxy-2-(3-methyl-2-butenyl)bibenzyl,(3)3-methoxy-5-hydroxy-2-(3-methyl-2-butenyl)bibenzyl,(4)quercetine(3,5,7,3′,4′-Pentahydroxy flavone),(5)5,7-Dihy droxyflavone,(6)methyol pinostrobin(5-hydroxy-7-methoxy-flavanone),(7)(7E)-7,8-Dehydroheliobuph thalmin,(8)3,4:3′,4′-Bis(methylenedioxy)-lign-7(E)-en-9,9′-olide,(9)3,4:3′,4′-Bis(methylenedioxy)-lign-7(Z)-en-9,9′-olide,(10)hinokiresinol(1,3-Double-(4-hydroxy phenyl)-1,4-pentadiene),(11)ginkgo phenol((Z)-3-(tridec-8-en-1-yl)phenol),(12)tree bean keto acid A(3-hydroxy-4-prenyl-5-methoxystilben-2-carboxylic acid).These above compounds include four flavonoids,two toluylenes,one phenol,three xylogens and two phenylpropanoids.Especially the Compound 1*,namely thuja phenol A,is a new compound.And these twelve compounds are all first found in Thuja plants.In this experiment,it was found that the alcohol extract of T.sutchuenensis plant is rich in triterpenoids and flavonoids,and is rich in bioactive compounds which are of great value.In addition to the new compound monoterpene substituted distyrene A(1*)has been published in this paper,several other new compounds have been possibly extracted,and the molecular structure is being further identified.Finally,the inhibitory activity of the new compound 1*(thuja phenol A)on four cancer cells and three bacteria were also tested.The cytotoxic activities of the compound 1* on non-small cell lung cancer cell and other three kinds of human tumor cells were not obvious(IC50 > 100 μmol/mL).And the antibacterial effects on the three kinds of bacteria such as MRSA / SA / Eco were effective(All MIC were 25μg/mL).
