| Objective:Analyze the effect of Alkaloid Sinomenine(SIN)in terms of sepsis rats’ TH1/TH2,endothelial and lung injury to discuss the application of SIN in treating Septicopyemia.Materials and methods: Choose inbred male rat Lewis and divide them into 4 groups,namely sham group,model group,model L-SIN group and H-SIN group with 8 rats in each group.Adopt randomized controlled trial to establish the model of Septicopyemia rats.Sham group:inject normal saline into caudal vein 0.5ml;Model group:inject normal saline into caudal vein 0.5ml.L-SIN group:inject Alkaloid Sinomenine solution into caudal vein 0.5ml(dosage: 50mg/kg);H-SIN group: inject Alkaloid Sinomenine solution into caudal vein 0.5ml(dosage: 100mg/kg).Kill the rats 24 hours later.Adopt ELISA method to detect the level of serum IL-2,TNF-α,IL-4 and IL-10.At the same time,measure the selectin of ICAM-1 、 VCAM-1 、 P-and adopt chromosomal HE to detect the pathology change of lung tissues and collect datum to conduct statistical analysis.Results: 1.After the operation,rat’s in Sham group are in good spirit with smooth hair;While rat’s in Model group,L-SIN group and H-SIN group are not in good spirit with rough hair.Eating and drinking are limited,especially for Model group.Rats in model group all have the symptom of shivering.L-SIN group has 7 examples,but H-SIN group only has 2 examples.After opening the abdominal cavity,rats in model group all has abscess.L-SIN group has 6 examples,H-SIN group has 2 examples.Sham group is normal.2、IL-2:Compared with model group,H-SIN group has significant differences,while L-SIN group does not have significant differences.TNF-α : Compared with sham group,L-SIN group has remarkable differences while H-SIN group does not have remarkable differences.Compared with model group,H-SIN group、L-SIN group have significant differences.IL-4: Compare H-SIN group and L-SIN group with sham group,noticeable difference can be discovered.Compared with model group,H-SIN group、L-SIN group have significant differences.Compare H-SIN group with L-SIN group,noticeable difference can be discovered.IL-10 :Compared with model group,H-SIN group has remarkable differences;Compare H-SIN group with L-SIN group,noticeable difference can be discovered.3.ICAM-1:Compare H-SIN group and L-SIN group with model group and sham group,noticeable difference can be discovered.Compared with model group,H-SIN group and L-SIN group have significant differences.VCAM-1:Compared with sham group,H-SIN group,L-SIN group and model group have significant differences.Compared with model group,H-SIN group、L-SIN group have remarkable differences.P-selectin:Compare Model group and L-SIN group with sham group,chromosome is greatly different;Compared with model group,H-SIN group has remarkable differences.4、After the HE staining of lung and heart tissues,sham group does not appear inflammatory cellular infiltration.The capillary enlarges and appears congestion in Model group and appears inflammatory cellular infiltration.A large number of alveolar epithelial has the symptoms of edema and degeneration.L-SIN group has the symptoms of edema and degeneration,and the capillary enlarges and appears congestion in Model group and appears tons of inflammatory cellular infiltration.H-SIN group has the symptoms of edema and degeneration,and the capillary enlarges and appears congestion in Model group and appears limited number of inflammatory cellular infiltration Conclusion:1.Alkaloid Sinomenine is able to regulate the cytokines of Septicopyemia’s TH1/TH2 and suppress the inflammatory reaction.2.Alkaloid Sinomenine is capable of suppressing the express of vascular endothelial factor to improve the function of vascular endothelial.3.Alkaloid Sinomenine can improve the lung damage caused by Septicopyemia and certain protection for multiple organ multifunction. |
