[Background and Objective]Hydrogen sulfide H2S),as a novel neuro-protectant,ameliorates postoperative cognitive dysfunction POCD).However,the mechanisms remain largely unknown.Warburg effect aerobic glycolysis)plays an important role in regulating learning and memory.2-Deoxy-D-glucose 2-DG)is a recognized inhibitor of Warburg effect.The present work explored the influences of H2 S on Warburg effect in the brain of postoperative aged rats and whether 2-DG inhibits H2S-exerted change in the warburg effect in the brain of postoperative aged rats.This will lay a strong foundation for our further study whether Warburg effect mediates the antagonistic effect of H2 S on POCD.[Methods]The expressions of Warburg effect-related protein,including Hexokinase II HK2),Pyruvate kinase M2 PKM2),Pyruvate dehydrogenase kinase 1 PDK1),Pyruvate dehydrogenase PDH),Lactate dehydrogenase A LDHA)in brain tissues were detected by western blotting.The content of lactate in brain tissues was detected using lactic acid kit.[Results]1.H2 S upregulated Warburg effect in the brain of postoperative aged rats.Sodium hydrosulfide Na HS,a donor of H2 S,100 ?mol/kg,i.p.)significantly increased the expressions of HK2,PKM2,LDHA,and PDK1,decreased the expression of PDH,and elevated the level of lactate in the brain of postoperative aged rats.These results indicated that H2 S ameliorates Warburg effect in the brain of postoperative aged rats.2.2-DG suppressed H2S-enhanced Warburg effect in the brain of postoperative aged rats2-DG 100 mg/kg,i.p.)markedly inhibited H2S-elicited upregulation in the expressions of HK2,PKM2,LDHA,and PDK1,downregulation in PDH expression,and increase in lactate content,in the brain of postoperative aged rats.These results indicated that 2-DG inhibits H2S-enhanced Warburg effect in the brain of postoperative aged rats.[Conclusion]1.H2 S enhances Warburg effect in the brain of postoperative aged rats;2.2-DG blocks the up-regulatory role of H2 S in Warburg effect in the brain of postoperative aged rats. |
