Objective:To establish a model of non-alcoholic fatty liver disease(NAFLD)rats,and to investigate the effects of ginsenoside Rgl on apoptosis of hepatocytes and its mechanism.Method:Forty SD rats were randomly assigned to four group(n=10),including control group,model group,Ginsenoside Rg1 low dose group(LD Rg1),Ginsenoside Rg1 high dose group(HD Rg1).The control group was fed with normal diet,the model group was fed with high-fat,high-sugar diet(HFHSD).The other groups including LD Rg1 group and HD Rgl group were fed with HFHSD for 14 weeks,then the two groups were treated with LD Rg1 and HD Rg1 group for 8 weeks respectively.Testing the serum to detect liver fuction and blood lipid.Collecting liver tissue to do HE for liver morphology observation and do oil red staining for watching liquid in liver tissue.Using TUNEL for detecting the degree of apoptosis.Taking the serum to detect oxidative stress index MDA,SOD,GSH as well as we take the liver tissue to test expression of the key apoptosis factors Bax,Bcl-2,Procaspase-3,Caspase-3 cleavage p17 through IHC,Western-blotting from protein level.Also,we detect the mRNA level of these factors in liver cells.Result:by detecting liver fuction index and bloody lipid level,we found out compared with the control group,the level of ALT and AST in the model control group increased,and the TC and TG also increased.Compared with the control group,the liver function index of ginsenoside Rgl was decreased,and the serum lipid level decreased gradually.Liver morphology had shown disordered arrangement of liver cells,large fat lipids,and inflammatory cells infiltration in model group,compared with control group.After treatment with Ginsenoside Rg1,the arrangement of liver cells got closed,fat lipids reduced.TUNEL stain shows that model group got much more small particles than control group,however,LD Rgl group and HD Rg1 group got lesser small particles than model group.In comparison with the control group,the outcomes from detecting oxidative stress indexes shows that MDA declined in model group(P<0.05)and upregulated after treated with Rg1(P<0.05).On the contrary,SOD and GSH elevated in the model group(P<0.05),after Rg1 treatment the trend was down regulating in the both factors(P<0.05).Compared with control group,the mRNA Transcription shows that BAX and Caspase-3 was promoted,but Bcl-2 downregulate in the model group,through treatment with the Ginsenoside Rgl,compared with model group,BAX and Caspase-3 decreased,Bc1-2 increased in both LD Rg1 group and HD Rg1 group.Meanwhile,protein expression of the key apoptotic indexes BAX,Caspase-3 cleavage p17 increased in the model group,reversely,the Bcl-2 and Procaspase-3 decreased,compared with control group.After treatment with Rgl for 8 weeks BAX,Caspase-3 cleavage p17 is down regulating and that Bcl-2,Procaspase-3 is up regulating in LD Rg1 group and HD Rg1 group.Conclusions:In NAFLD,oxidative stress induced apoptosis was one of the main factors causing liver injury.Ginsenoside Rgl has the effect of improving the liver injury of NAFLD,and its mechanism may be related to the inhibition of liver cell apoptosis by alleviating oxidative stress in the liver.
